摘  要:目的探讨强志胶囊对抑郁模型大鼠神经行为及不同脑区单胺类神经递质的影响。方法健康雄性Wistar大鼠(SPF级)60只,采用慢性温和应激法(CMS)制备抑郁大鼠模型,适应性饲养一周后据旷场实验及糖水偏嗜实验基数期数据随机分成5组,灌胃给药,每日一次。以体质量检测大鼠生理代谢情况,糖水偏嗜实验检测模型大鼠兴趣缺失情况,旷场实验检测运动和探究行为情况;Elisa检测CRH、ACTH、CORT等HPA轴激素含量;ELISA检测海马、下丘脑、额叶皮层5-HT、NE、DA含量。结果行为参数方面:相较于模型组,强志胶囊的大、小剂量组体质量、糖水偏嗜系数及旷场总路程、中央区路程、直立次数均显著升高(P<0.001)。神经生化指标方面:相较于模型组,强志胶囊大、小剂量组大鼠血清中CRH、ACTH含量均显著降低(P<0.01,P<0.001),CORT含量无显著变化。单胺神经递质方面:5-HT在额叶皮层含量显著降低(P<0.001);DA在额叶、下丘脑、海马三个脑区均显著性降低(P<0.001);NE在额叶皮层内含量显著降低(P<0.05),在下丘脑内含量升高(P<0.05)。结论强志胶囊能显著改善抑郁模型大鼠情绪异常行为,其作用可能与强志胶囊可以调节HPA轴功能,影响中枢单胺神经递质含量相关。Objective To study the effect of Qiang Zhi Capsule on neurobehavior and monoamine neurotransmitters in different brain regions of depression model rats. Methods Depression model was established by chronic mild stress method(CMS). Sixty healthy male Wistar rats(SPF grade) were conventional raised a week, then were randomly divided into 5 groups, administration by gavage, once daily. Depressive-like behaviors were tested by Body weight, sucrose preference experiment and open field;Content of CRH, ACTH, CORT in serum and 5-HT, NE and DA in hippocampus, hypothalamus and frontal cortex were tested by ELISA Kit.Results Compared with the model group, the body weight, sucrose preference coefficient, total distance, the central area distance and straight times were significantly higher in the large and small dosage group of Qiang Zhi Capsule(P<0.001). In terms of related biochemical indexes: Compared with the model group, the contents of CRH and ACTH in the high and low dose groups of Qiang Zhi Capsule were significantly reduced(P<0.01, P<0.001), and the contents of CORT were not significantly changed. In terms of monoamine neurotransmitter, 5-HT decreased significantly in frontal cortex(P<0.001);DA decreased significantly in frontal, hypothalamus and hippocampus(P<0.001);NE decreased significantly in frontal cortex(P<0.05), and increased in hypothalamus(P<0.05). Conclusion Qiang Zhi Capsule can significantly improve the abnormal behavior of depression model rats, and its effect may be related to that Qiang Zhi Capsule can regulate HPA axis function and affect the content of Central Monoamine Neurotransmitter.
