摘 要:目的分析枸杞多糖(Lycium barbarum polysaccharide, LBP)对2型糖尿病(Type 2 diabetes mellitus, T2DM)大鼠外周血、肾脏、脾脏髓源抑制性细胞(myeloid-derived suppressor cells, MDSCs)及血浆炎症因子的影响。方法 20只SD大鼠,高脂饲料喂养联合腹腔注射35mg/kg链脲佐菌素(STZ)构建2型糖尿病大鼠模型。造模成功后随机分为T2DM组与LBP干预组,每组10只。干预8周后,流式细胞术检测各组外周血、肾脏与脾脏中MDSCs比例变化,大鼠CBA炎症细胞因子试剂盒分析各组血浆中IL-10、IL-17A、TNF-α与IL-6的水平,Pearson分析两者相关性。结果与T2DM组相比,LBP干预组大鼠外周血、肾脏及脾脏中的MDSCs比例显著增加(P<0.05),血浆TNF-α、IL-17A与IL-6表达水平降低(P<0.05),IL-10升高但差异无统计学意义(P>0.05)。分析外周血、肾脏、脾脏细胞中MDSCs比例与炎症因子相关性发现,MDSCs与IL-17A、TNF-α及IL-6呈负相关。结论 LBP可诱导MDSCs在外周血、肾脏和脾脏中的募集,减少促炎因子IL-17A、TNF-α和IL-6的生成,发挥抗炎作用,延缓T2DM的进展。Objective To investigate the effects of lycium barbarum polysaccharide(LBP) on myeloid-derived suppressor cells(MDSCs) in peripheral blood, kidney, spleen and plasma inflammatory cytokines in rats with type 2 diabetes mellitus(T2 DM).Methods T2 DM model was induced by high-fat diet in combination with intraperitoneal injection of 35 mg/kg streptozotin(STZ). All rats were randomly divided into two groups: diabetes group(T2 DM) and LBP intervention group(T2 DM+LBP). And then the levels of MDSCs from peripheral blood, kidney, and spleen were detected by flow cytometry after 8 weeks of intvervention, respectively. The levels of IL-10,IL-17 A,TNF-α and IL-6 in plasma were measured by flow cytometry CBA. Statistical Pearson was used to analyze the correlation between MDSCs and inflammatory cytokines. Results Compared to T2 DM group, the proportions of MDSCs from peripheral blood, kidney and spleen in T2 DM+LBP group were significantly increased, respectively( P<0.05). The levels of plasma IL-17 A, TNF-α and IL-6 in T2 DM+LBP group were lower than those in T2 DM group, respectively( P<0.05). There’s no significant difference of IL-10 between two groups( P>0.05). The percentage of MDSCs in peripheral blood, kidney and spleen were negatively correlated with the levels of IL-17 A, TNF-α and IL-6, respectively. Conclusion LBP may contribute to suppressing inflammation via recruiting MDSCs and reducing the levels of pro-inflammatory cytokines, thereby alleviating the progression of the disease.
