摘 要:目的研究弥散吸氧对移居高原人员缺氧应激、血氧饱和度以及人体代谢过程的影响,阐明新型供氧方式的抗缺氧作用及其可能的分子机制。方法以移居海拔3540 m高原某地1年以上的10名高原人员为研究对象,以新研发的弥散供氧住宿方舱为实验场所,研究对象在住宿方舱内连续弥散吸氧3 d,2 h/d。采用自身对照研究方法,检测研究对象吸氧前后生理指标、血常规指标及血浆代谢谱的变化。结果(1)研究对象在住宿方舱内弥散吸氧期间,第1天舱内氧浓度为(22.85±0.36)%、第2天为(22.52±0.76)%、第3天为(22.57±1.05)%。(2)研究对象吸氧后血氧饱和度显著提高、脉率明显下降(P<0.01);血压吸氧前后比较差异无统计学意义。(3)研究对象吸氧后共有44种代谢物发生了显著改变(VIP>1,P<0.05)。其中,正离子模式下鉴定出18种,负离子模式下鉴定出26种。发现主要有4条代谢通路发生了明显变化[impact>0.05,-log_(10)(P)>1.3]。结论通过弥散供氧方式吸氧能显著提高机体的心肺供氧能力,有效缓解高原缺氧寒冷环境对人体代谢的影响。Objective To determine the effects of diffuse oxygen inhalation on hypoxia stress,blood oxygen saturation and human metabolic process in migrants in plateau,and to illustrate the anti-hypoxia effect of a new oxygen supply mode and its possible molecular mechanism.Methods Ten workers who had migrated to a plateau at an altitude of 3540 m for more than 1 year were recruited as the subjects.They were asked to breathe oxygen in a newly developed accommodation cabin with diffuse oxygen supply for 3 consecutive days,2 h per day.Their physiological indexes,blood routine indexes and plasma metabolic profiles before and after oxygen intake were measured and compared.Results(1)The oxygen concentration was(22.85±0.36)%on the first day,(22.52±0.76)%on the second day and(22.57±1.05)%on the third day during the diffusion oxygen inhalation in the accommodation cabin.(2)The oxygen saturation level and pulse rate were significantly increased after oxygen inhalation in the subjects(P<0.01),but there was no significant change in blood pressure(P>0.05).(3)There were 44 metabolites significantly changed(VIP>1,P<0.05).Among them,18 metabolites were identified in the positive ion mode and 27 in the negative ion mode.And 4 major metabolic pathways also had significant changes[impact>0.05,-log_(10)(P)>1.3].Conclusion Oxygen inhalation via diffuse oxygen supply can effectively improve the capacity of cardiopulmonary oxygen supply,and alleviate the impact of hypoxia and cold environment on human metabolism.
