摘 要:目的通过对10批次地黄主产区所产的生地黄煅炭后的高效液相色谱(HPLC)指纹图谱研究,结合《中国药典》中规定的质量检测方法,建立生地黄煅炭品的指纹图谱分析方法,为有效评价和控制其质量提供依据。方法取地黄煅炭粉末,用甲醇回流提取,以乙腈-0.1%磷酸水为流动相,梯度洗脱,检测波长为210.0nm,柱温30℃,流速1mL/min。采用"中药色谱指纹图谱相似度评价系统"软件(2004A版,国家药典委员会编)进行数据处理分析,计算不同产地生地黄煅炭的相似度。结果确定在波长为210.0 nm处生地黄煅炭指纹图谱有17个共有峰,不同产地生地黄煅炭之间的相似度均大于0.8,除山西1外。结论该研究建立的生地黄煅炭指纹图谱分析方法精密度、重复性和稳定性良好,可有效用于生地黄煅炭的质量评价。Objective The content of the main active components in 10 batches of calcined charcoal of Rehmannia glutinosa was determined by HPLC, which provided an effective method for controlling the quality of calcined charcoal of Rehmannia glutinosa. Methods The calcined carbon powder of Rehmannia glutinosa was extracted by methanol reflux, acetonitrile 0.1% phosphoric acid aqueous solution was used as mobile phase. The method of gradient elution was used to detect the wavelength of 210.0 nm, column temperature was 30 degrees centigrade, flow rate of 1 mL/min. The similarity evaluation software(2004 A version) of traditional Chinese medicine chromatographic fingerprint prepared by pharmacopoeia committee was used for data processing, and the similarity between the extracts of calcined charcoal of Radix Rehmanniae from five habitats was calculated by the angle cosine method. Results It was determined that there were 17 common peaks in fingerprint of calcined charcoal of Rehmannia glutinosa under the wavelength of 210 nm, and the similarity between calcined charcoal of Rehmannia glutinosa from different origin was greater than 0.8, except for shanxi 1. Conclusion The fingerprint of alcohol extracts of Rehmannia, the established method is accurate and reliable, reproducible and can be used to evaluate the different origin of Rehmannia root quality;and can be used as the standard, to provide the basis for further quality control of calcined charcoal of Rehmannia glutinosa.
