摘 要:目的该研究以红景天药材为例,将具有中药特色的系统生药学鉴定技术应用于红景天的鉴定,为临床用药安全提供依据。方法基于药典方法对来源于8个产地的红景天进行性状、显微、薄层色谱鉴定;采用DNA条形码技术,通过ITS1序列比对、遗传距离比较和系统NJ树构建进行DNA分子鉴定。结果红景天的性状鉴定、显微鉴定、理化鉴定结果可以作为红景天鉴定的依据,但不能有效评价其真伪;红景天样本的ITS1序列长度为226 bp,种间最大遗传距离为0.06,系统发育树图中23个样本为5个品种红景天聚为两支,正品大花红景天容易区分。结论基于系统生药学鉴定鉴定方法能够准确、有效的鉴定红景天药材的真伪。Objective Authenticity identification of Chinese medicine R.crenulata and its adulterants was performed by systematic pharmacognosy identification combining DNA barcoding technology with traditional macroscopical characteristics, microscopical characteristics and physicochemical identification technology, to ensure the quality and safety of Chinese medicinal materials. Methods Commercially available Rhodiola from eight producing areas were identified by macroscopical characteristics, microscopical characteristics and thin layer chromatography based on pharmacopoeia. ITS1 and ITS2 sequence were employed for DNA barcoding, and amplified by PCR, K2 P distances were calculated and NJ tree was performed. Results The results of morphological characteristics, microscopical characteristics and thin layer chromatography identification could not effectively identified the authenticity of Rhodiola, and interspecific K2 P distance of ITS1 was 0.06, and ITS1 sequence of R.crenulata and other 4 species could be well distinguished by NJ tree. Conclusion Systematic pharmacognosy Identification could efficiently, and accurately identify the authenticity of Rhodiola specifically.
