摘 要:为确定海带(Saccharina japonica)配子体紫外线辐射的适合剂量范围,利用紫外线对东方海洋种质库保存的2个海带配子体克隆系进行辐射试验。通过三次预试验,辐射条件设置为:辐射距离12 cm,辐射时间分别为10、20、30、40、50、60、90、120 s。结果显示:辐射时间10、20 s配子体细胞未发生明显变化,但受辐射影响,克隆约2个月的时间不生长;辐射时间30 s细胞存活率约为50%~55%;随着辐射时间的增加,细胞死亡增多;辐射时间120 s下,两个克隆系只有极个别细胞存活。在辐射距离12 cm时,辐射时间10~120 s作用范围内,雄配子体耐受力明显高于雌配子体。Four ultraviolet radiation experiments were carried out on two gametophyte clones of Saccharina japonica,preserved in the germplasm bank of Shandong Oriental Ocean Sci-tech Co.Ltd.to screen the suitable dose range of ultraviolet radiation for mutation breeding.After three pre-tests,the experiment was set 12 cm for radiation distance,and 10 s,20 s,30 s,40 s,50 s,60 s,90 s,120 s for radiation time,respectively.The results showed that no cell survived when the radiation distance was 6 cm.The gametophyte cell did not change significantly for 10 s and 20 s when the radiation distance was 12 cm,but the clones did not grow during a period of approximately 2 months;the survival rates of clones were about 50%~55%for 30 s;the number of death cells increased with the increase of radiation time;only a few cells survived for 120 s.The tolerance of male gametophytes was significantly stronger than that of female gametophytes when the radiation distance was 12 cm and the radiation time was 10~120 s.
