摘  要:以毛豆为研究对象,采用超声波辅助水提取法对其进行提取,通过ABTS+·清除能力测试、DPPH·清除能力测试、胶原蛋白酶抑制能力测试、弹性蛋白酶抑制能力测试和晚期糖基化终产物AGEs的清除能力测试等方法,对毛豆提取物的体外抗衰老活性进行考察评估。结果显示,毛豆提取物具有良好的体外抗氧化和抗糖化活性:清除ABTS+·的IC_(50)值为11.16 mg/mL,清除DPPH·的IC_(50)值为11.77 mg/mL,清除AGEs的IC_(50)值为3.21 mg/mL;对与皮肤衰老紧密相关的多种蛋白酶显示出很好的体外抑制活性:抑制胶原蛋白酶的IC_(50)值为12.91 mg/mL,50mg/mL的毛豆提取物具有与阳性对照药物西维来司钠(50μmol/L)相当的抑制弹性蛋白酶的能力。以上结果表明,毛豆提取物具有较好的体外抗衰老活性。In this study, soybean was extracted by ultrasonic assisted extraction. The in vitro anti-aging activity of the soybean extracts was tested by ABTS+· scavenging ability test, DPPH· scavenging ability test, collagen protease inhibition test, elastase inhibition test and advanced glycation end products(AGEs)scavenging ability test. The results suggested that the soybean extracts displayed significant antioxidant and anti-glycation activities in vitro: the IC_(50) for ABTS+· scavenging was 11.16 mg/mL, the IC_(50) for DPPH· scavenging was 11.77 mg/mL, and the IC_(50) of AGEs scavenging was 3.21 mg/mL;The extracts also showed great inhibitory activity against various proteases related to skin aging: the IC_(50) of collagenase inhibition was 12.91 mg/mL, and 50 mg/mL of soybean extracts had similar inhibitory effects on elastase as the positive control(Civeilex sodium, 50 μmol/L). These results indicated that the soybean extracts have good anti-aging activity in vitro.
