摘  要:目的:探究噪声性耳聋大鼠耳蜗组织中miRNA-183表达情况,并分析其表达水平变化对噪声性耳聋发生发展的影响。方法:将50只SD大鼠分为两组,其中10只设为空白对照组,另40只建立噪声性耳聋动物模型,并随机分为噪声暴露1 d组(n=10)、噪声暴露7 d组(n=10)、噪声暴露14 d组(n=10)、噪声暴露28 d组(n=10)。处死各组大鼠取出耳蜗组织,应用生物显微镜观察耳蜗基底膜内外毛细胞排列情况,并应用RT-PCR法检测miRNA-183表达水平,比较各组大鼠耳蜗基底膜镜检结果及miRNA-183表达差异。结果:在生物显微镜下可见,空白对照组大鼠耳蜗基底膜的内毛、外毛细胞列整齐,而噪声暴露1 d组、噪声暴露7 d组、噪声暴露14 d组、噪声暴露28 d组的外毛细胞排列均紊乱。Tamhane事后检验显示,外毛细胞缺失率随噪声暴露时间延长而显著升高(P<0.05)。与空白对照组相比,噪声暴露1 d组、噪声暴露7 d组、噪声暴露14 d组、噪声暴露28 d组的miRNA-183相对表达量均显著降低(均P<0.05)。结论:miRNA-183在噪声性耳聋大鼠耳蜗组织中呈低表达,miRNA-183表达量下调可能会促进噪声性耳聋发生发展,而且对耳蜗毛细胞损伤修复评估具有一定的临床意义。Objective:To investigate the expression of miRNA-183 in the cochlea tissue of noise-induced deafness rats,and analyze the influence of its expression level on the development of noise-induced deafness.Methods:Fifty SD rats were divided into two groups,of which 10 SD rats were set as blank control group,and another 40 SD rats were established as animal models of noise-induced deafness,and randomly divided into noise exposure 1 day group(n=10),Noise exposure 7 days group(n=10),noise exposure 14 days group(n=10),noise exposure 28 days group(n=10).The rats in each group were sacrificed and the cochlear tissues were taken out.The arrangement of the inner and outer hair cells of the cochlear basement membrane was observed with biological microscope,and the expression level of miRNA-183 was detected by RT-PCR.The results of the cochlear basement membrane microscopy and miRNA-183 expression of each group were compared.Results:It can be seen under the biological microscope that the inner and outer hair cells of the cochlea basement membrane of the blank control group were neatly arranged,while the outer hair cells of the other four groups were arranged disorderly.The loss rate of outer hair increased significantly with prolonged noise exposure time(P<0.05).Compared with the blank control group,the relative expression of miRNA-183 in the other four groups was significantly reduced(all P<0.05).Conclusion:The expression of miRNA-183 is low in the cochlear tissue of noise-deaf rats.Down-regulation of miRNA-183 expression may promote the occurrence and development of noise-deaf,and has certain clinical significance for the evaluation of cochlear hair cell damage repair.
