摘  要:为了研究牛防御素5(BNBD5)微球对卡介苗(BCG)免疫小鼠的影响,在表达纯化获得目的蛋白BNBD5后,制备包封BNBD5重组蛋白的聚乳酸-羟基乙酸(PLGA)纳米微球,并进行动物试验。用BCG对BALB/c小鼠免疫后,再用BNBD5纳米微粒对小鼠进行滴鼻,通过ELISA方法检测小鼠血清中细胞因子和抗体的变化,发现BNBD5-PLGA微球能够显著提高促炎因子IL-β(P<0.001)和TNF-α(P<0.001)以及抗体IgA(P<0.001)产生;再用牛分枝杆菌(Mycobacterium bovis)滴鼻感染小鼠,发现BNBD5-PLGA组临床症状和病理变化有所减轻。表明重组蛋白牛防御素5微球能刺激小鼠产生较高的非特异性细胞因子以及黏膜免疫抗体IgA水平,对黏膜免疫具有促进作用。To study the effect of bovine neutrophilsβ-defensins 5(BNBD5)nanoparticles on bacilli-calmette-guerin(BCG)immunized mice,we prepared the BNBD5-PLGA nanoparticle complexes and conducted animal experiments.After immunizing BALB/c mice with BCG,the mice were dripped with BNBD5 nanoparticles.From the results of cytokines and antibodies in the serum or lung lavage of mice by ELISA,we found that the BNBD5-PLGA microspheres significantly improved the production of pro-inflammatory factors IL-β(P<0.001),TNF-α(P<0.001)and IgA(P<0.001).The immunized mice were then challenged with Mycobacterium bovis.Clinical symptoms and pathological changes were relieved in the BNBD5-PLGA group.These data confirm that recombinant protein bovine defense 5 microspheres can stimulate mice to produce higher nonspecific cytokines and mucosal IgA levels,suggesting that it has a catalytic effect on mucosa immunity.
