摘  要:目的明确十堰地区腊梅花精油的成分并在大鼠弗氏完全佐剂致足肿胀模型中探索其抗炎作用,为腊梅花资源的进一步开发提供依据。方法将SD大鼠随机分为空白对照组、模型组、阳性对照组(阿司匹林200 mg/kg)及腊梅花精油低、中、高剂量组(0.1、0.3、0.8 g/kg),灌胃给予相应药物或溶剂,给药容积10 mL/kg,1次/d。于第5天给药后30 min,大鼠右侧足跖皮下注射弗氏完全佐剂0.1 mL,空白对照组给予生理盐水0.1 mL。观察并用排水法测量致炎前后1、2、3、5、24、48、72 h足趾体积,计算各时间点足趾肿胀率,并用酶联免疫吸附试剂盒检测足部肿胀组织中炎症因子的水平。结果腊梅花精油中、高剂量组对弗氏完全佐剂致炎大鼠1、2、3、5、24、48 h的足趾肿胀均明显抑制(P<0.05),各剂量组肿胀足部组织中的TNF-α和IL-1β的含量明显减少。结论腊梅花精油对弗氏完全佐剂致大鼠足趾肿胀有明显的抑制作用,其抗炎作用的发挥可能与抑制TNF-α和IL-1β有关。Objective To clarify the composition of essential oil extracted from Chimonanthus praecox flowers in Shiyan area and to explore its anti-inflammatory effects to provide support for further development of the resources of Chimonanthus praecox flowers.Methods SD rats were randomly divided into blank control group,model group,positive control group(aspirin 200 mg/kg),and low-,medium-and high-dose essential oil groups(0.1,0.3 and 0.8g/kg).The blank control group and the model group were treated with distilled water by intragastric administration.The positive control group was treated with aspirin by intragastric administration.The low,medium,and high doses of the essential oil extracted from Chimonanthus praecox flowers were given 10,30 and 80 mL/L at 10 mL/kg once a day.On day 5 of the experiment,30 minutes after intragastric administration,0.1 mL of Freund’s complete adjuvant was injected subcutaneously into the right foot plantar of each group of rats,and the blank control group was subcutaneously injected with 0.1 mL of normal saline.We observed and measured the toe’s volume of the rats before and 1,2,3,5,24,48,and 72 h after injection by using drainage method.We then calculated the toe’s swelling rate in each group of rats at each time point,and used ELISA kit to measure the content of inflammatory factors in swollen foot tissue.Results In the medium-and high-dose essential oil groups,we observed significant inhibitory effects on the toe’s swelling rate in rats at 1,2,3,5,24,and 48 h after inflammation with Freund’s complete adjuvant(P<0.05).The essential oil extracted from Chimonanthus praecox flowers could significantly decrease the contents of TNF-αand IL-1βin the swollen foot tissue,and its anti-inflammatory effect was dose-dependent.Conclusion The essential oil extracted from Chimonanthus praecox flowers has obvious inhibitory effects on the rate of the toe’s swelling induced by Freund’s complete adjuvant.The anti-inflammatory effect may be related to the inhibition of TNF-αand IL-1β,but i
