摘 要:为分析江苏地区仔猪腹泻性大肠杆菌的致病性、血清型、耐药性及生物被膜(BF)形成能力等生物特性,本研究从2017年~2019年采集江苏地区患腹泻的仔猪粪便、肛拭子等216份病料样品,从中分离培养后经16S rRNA PCR鉴定获得156株大肠杆菌,进一步通过小鼠感染性实验得到118株致病性大肠杆菌。对分离的致病性大肠杆菌采用O血清型玻板凝集试验检测其血清型,结果显示118株致病性大肠杆菌有14种血清型,以O38型、O149型及O109型为主要的优势流行血清型;采用K-B药敏纸片法对分离菌株的耐药性检测结果显示,118株致病性大肠杆菌对氨苄西林、磺胺间甲氧嘧啶、环丙沙星等7种药物的耐药率在47.5%以上,对其他药物耐药率在11.0%~16.1%;结晶紫微孔板法检测分离菌BF的形成能力,结果显示,分离的118株致病性大肠杆菌中BF形成能力强、中等、较弱及不形成BF的菌株分别有48株(40.7%)、36株(30.5%)、24株(20.3%)、10株(8.5%),且对氨苄西林、磺胺间甲氧嘧啶、环丙沙星等7种药物的耐药性与BF形成能力具有一定相关性。本研究为仔猪腹泻性大肠杆菌病的防控提供科学依据。In order to analyze the pathogenicity,serotype,drug resistance and biofilm forming ability of diarrheagenic E.coli in piglets in Jiangsu province,156 E.coli strains were isolated from 216 samples of piglet feces,anal swabs and other diseased tissues from farms in Jiangsu province from 2017 to 2019 in this study.Then,118 strains of pathogenic E.coli were obtained through the infectivity test in mice.The serotypes were detected by glass slides agglutination test,and the results showed that 118 strains of pathogenic E.coli belonged to 14 serotypes,with O38,O149 and O109 as the dominant serotypes.The drug resistance of the isolated strains was detected by K-B drug sensitivity disk method,and the results showed that the drug resistance rate of 118 pathogenic E.coli strains to ampicillin,sulfamethoxine and ciprofloxacin was over 47.5%,and to other drugs was 11.0%-16.1%.The ability of biofilm formation was detected by crystal violet microplate method,and it was shown that 48 strains(40.7%),36 strains(30.5%),24 strains(20.3%)and 10 strains(8.5%)were found to have strong,medium,weak biofilm forming ability and no biofilm forming ability,respectively.Additionally,the drug resistance to 7 kinds of drugs,such as ampicillin,sulfamethoxine and ciprofloxacin,was correlated to biofilm forming ability.This study provides scientific basis for prevention and control of colibacillosis-related diarrhea in piglets.
