摘 要:伯克霍尔德氏菌(Burkholderia sp.)YZU-S230是长江大学植物与微生物互作研究室前期分离的一株白芨内生细菌,对水稻纹枯病菌(Rhizoctonia solani)、稻瘟病菌(Magnaporthe grisea)、西瓜枯萎病菌(Fusarium oxysporum f.sp.niveum)、马铃薯晚疫病菌(Phytophthora infestans(Mont)de Bary)、草莓灰霉病菌(Botrytis cinerea)、棉花枯萎病菌(Fusarium oxysporum f.sp.vasinfectum)等具有较强的抗菌活性。为筛选用于防治西瓜枯萎病的拮抗细菌,通过对峙培养、盆栽试验和绿色荧光标记测定了伯克霍尔德氏菌YZU-S230对西瓜枯萎病的防效和促生作用以及在西瓜体内的定殖动态。结果显示,YZU-S230对西瓜枯萎病菌(Fusarium oxysporum f.sp.niveum)的抑制率达到82.80%,且在温室条件下对西瓜枯萎病具有较好的防效,同时接种时其防效为30.87%,提前3d接种时其防效为68.40%。经YZU-S230处理后30d,西瓜主蔓长增加了8.7cm,根长增加了3.0cm,地上鲜重增加了6.9g,显著高于对照组。菌株YZU-S230能在西瓜根部稳定定殖,接种后1、3、5、7、9、15、21d其在西瓜根部的定殖量分别为80×10^(5)、120×10^(5)、29×10^(5)、9×10^(5)、1.7×10^(5)、1.4×10^(5)、1.2×10^(5)cfu/g。结果表明,菌株YZU-S230在防治西瓜枯萎病方面具有良好的开发应用潜力。Burkholderia sp.YZU-S230 is an endophytic bacterium of Bletilla striata isolated from plant and microorganism interaction in the early stage of the laboratory at Yangtze University.It has strong antifungal activity against Rhizoctonia solani,Magnaporthe grisea,Fusarium oxysporum f.sp.niveum,Phytophthora infestans(Mont)de Bary,Botrytis cinerea,Fusarium oxysporum f.sp.Vasinfectum,etc.To screen antagonistic bacteria for the control of watermelon Fusarium wilt,the control and growth-promoting effects of Burkholderia sp.YZU-S230 on watermelon Fusarium wilt and its colonization dynamics in watermelon were determined by confrontation culture,pot experiment and green fluorescence labeling.The results show that the inhibition rate of YZU-S230 to Fusarium oxysporum f.sp.niveum reaches 82.80%,and it has a good control effect on watermelon Fusarium wilt under greenhouse conditions with its control effect reaching 30.87%when inoculated simultaneously and 68.40%when inoculated 3d in advance.After 30 days of treatment with YZU-S230,the main vine length,the root length and aboveground fresh weight of watermelon increase by 8.7cm,3.0cm and 6.9g respectively,significantly higher than those of the control group.The strain of YZU-S230 could colonize stably in the root of watermelon,and the colonization amount at 1d,3d,5d,7d,9d,15d and 21d after inoculation are 80×10^(5),120×10^(5),29×10^(5),9×10^(5),1.7×10^(5),1.4×10^(5) cfu/g and 1.2×10^(5) cfu/g on,respectively.The results show that strain YZU-S230 has a good potential in controlling watermelon Fusarium wilt.
