摘  要:查尔酮合成酶(chalcone synthase,CHS)是类黄酮合成过程中的第一个关键酶,具有重要的生理功能。测定胡萝卜不同发育时期、不同组织部位的类黄酮含量,并在全基因组水平上对胡萝卜CHS基因家族进行鉴定和分析。结果表明,无论在苗期还是收获期,胡萝卜叶中的类黄酮含量均极显著地高于根中的含量。叶中的类黄酮含量在不同发育时期无显著差异,而根中的类黄酮含量在不同发育时期却差异显著。胡萝卜基因组中共有12个DcCHS基因,分布于5条染色体上,氨基酸序列长度为253~398 aa,外显子1~3个。经预测,DcCHS蛋白均定位于细胞质,二级结构以α-螺旋和无规卷曲为主。DcCHS蛋白可分为ClassⅠ和ClassⅡ两类,含有10种保守基序,motif1、4、6、7为CHS蛋白N端结构域,motif2、3、8为CHS蛋白C端结构域。这些研究结果可为胡萝卜CHS基因的功能研究提供参考。Chalcone synthase(CHS)is the first key enzyme in the synthesis of flavonoids and has important physiological functions.Flavonoids content of carrot was determined in different tissues at different developmental stages,and the CHS gene family was identified and analyzed from the complete genome of carrot.The results showed that the content of flavonoids in leaves was very significantly higher than that in roots at both seedling and harvest stages;there wasn’t significant difference inflavonoids content between different developmental stages,but flavonoids content of roots at seedling stage was significantly different from that at harvest stage.Twelve CHS genes were identified in the carrot genome,which distributed on 5 chromosomes.The length of DcCHS proteins ranged from 253 to 398 amino acids.The number of exons within DcCHS genes was 1-3.It was predicted that the secondary structure of DcCHS contained mainly alpha helix and random coil,and all the DcCHS proteins were located in cytoplasm.Ten motifs were identified in DcCHS proteins which were classified into two groups.motif1,4,6 and 7 were annotated as N-terminal domain,and motif2,3 and 8 were C-terminal domain.This study provides some useful evidences for further analyzing the functionof carrot CHS genes.
