不同代数间充质干细胞上清液对小鼠皮肤的影响

摘  要:目的探讨不同浓度间充质干细胞(mesenchymal stem cells,MSCs)上清液与不同代数MSCs对小鼠皮肤的影响。方法自2019年10月至2020年6月,郑州大学整形美容研究所通过体外培养从人脐带中剥离脐带MSCs,并进行传代收集。将其上清液提取物根据浓度和干细胞代数的不同分别注射于40只小鼠背部的4个象限位置,2次/周,6周后将小鼠处死,取皮肤组织行HE染色、维多利亚蓝染色、Masson染色,并使用Elisa法检测小鼠皮肤中的超氧化物歧化酶(superoxide dismutase,SOD)和丙二醛(malondialdehyde,MDA)含量。结果各组切片染色均无明显病理变化,注射MSCs上清液后会出现胶原纤维增加的变化,注射MSCs上清液的小鼠皮肤中MDA含量下降,SOD活力上升,MDA含量在注射P3代高浓度MSCs上清液时最低,SOD活力在注射P3代MSCs上清液时活力最高。结论在正常皮肤中注射MSCs上清液,可以引起其中胶原纤维和弹力纤维的变化,同时降低皮肤中的MDA含量,上调SOD的活性,起到抗衰老的作用。而MSCs传代至P3代的效果最为优异,且高浓度MSCs上清液的作用优于低浓度MSCs上清液。Objective To explore the effect ofdifferent concentrationsof mesenchymal stem cells(MSCs)supernatant and different generations of MSCs on skin of mice.Methods From October 2019 to June 2020,the umbilical cord MSCs extracted from human umbilical cord were cultured in vitro in the Plastic Surgery Institute of Zhengzhou University.The MSCs were collected after passage.According to different concentrations and generationsof MSCs,the supernatant extract was injected into 4 quadrants of the back of 40 mice twice a week.The mice were killedafter 6 weeks.The skin tissues were under went HE staining,Victoria blue staining and Masson staining.The content of superoxide dismutase(SOD)and malondialdehyde(MDA)in skin of mice was detected by ELISA.Results There were no obvious pathological changes in staining in each group.The collagen fiber increased after injection of MSCs supernatant.The activity of SOD increased.The content of MDA was the lowest when injection of high concentrations of MSCs supernatant of P3 generation,and the SOD activity was the highest.Conclusion The injection of MSCs supernatant in normal skin can induce the changes of collagen fiber and elastic fiber,reduce the content of MDA in the skinand up-regulate the activity of SOD,so as to play an anti-aging role.The MSCs of P3 generation had the best efficacy,and the performance of MSCs supernatant of high concentration is better than that of low concentration.
