摘  要:目的:基于肠道L细胞的葡萄糖转运体表达调控探讨小檗碱降糖机制。方法:以CCK8法及实时无标记细胞分析技术(RTCA)测定小檗碱对STC-1细胞活性的影响。ELISA及葡萄糖氧化酶(GOD-POD)法测定小檗碱对STC-1细胞胰高血糖素样肽-1(GLP-1)分泌及葡萄糖消耗的影响,利用qRT-PCR及Western blot技术分析细胞中钠/葡萄糖协同转运蛋白1(SGLT1)、葡萄糖转运蛋白2(GLUT2)mRNA及蛋白表达情况。结果:与正常组比较,5、10、50μmol/L小檗碱显著促进STC-1细胞分泌GLP-1(P<0.01,P<0.05)。10、50μmol/L小檗碱促进葡萄糖的消耗(P<0.01),上调STC-1细胞的SGLT1 mRNA表达(P<0.05),50μmol/L小檗碱上调SGLT1蛋白表达(P<0.05),而各浓度小檗碱对STC-1细胞的GLUT2 mRNA和蛋白的表达量无显著影响。结论:小檗碱促进STC-1细胞GLP-1分泌、提高糖消耗,其作用机制可能与上调SGLT1的表达有关。Objective:To explore the hypoglycemic mechanism of berberine based on the regulation of glucose transporter expression in intestinal L cells.Methods:The effects of berberine on the activity in STC-1 cells were determined by CCK8 and real-time cell analysis(RTCA).The effect of different concentration berberine on glucose consumption of STC-1 cells by glucose oxidase(GOD-POD)method and the level of GLP-1 was tested by ELISA.Western blot and qRT-PCR were used to evaluate the mRNA and protein expression of SGLT1 and GLUT2 in STC-1 cells.Results:Compared with the normal group,5,10,50μmol/L berberine improved the secretion of GLP-1 in STC-1 cells(P<0.01,P<0.05).10,50μmol/L berberine increased the consumption of glucose(P<0.01)and up-regulated the mRNA expression level of SGLT1(P<0.05).50μmol/L berberine upregulated the protein level of SGLT1(P<0.05).However,berberine at different concentrations has no significant effect on the upregulation of GLUT2 mRNA and GLUT2 protein expression in STC-1 cells.Conclusion:Berberine increase the GLP-1 secretion and glucose consumption,which mechanism may be related to the up-regulation of SGLT1 expression.
