摘 要:目的:探究不同初始肠道菌群对解毒祛瘀滋阴方治疗狼疮的影响。方法:雌性MRL/lpr狼疮小鼠随机分为模型对照组(灌胃0.9%氯化钠溶液,持续21d)、抗生素组(先灌胃抗生素混悬液14d,再灌胃0.9%氯化钠溶液7d)和粪菌移植组(先灌胃抗生素混悬液14d,再灌胃C57BL/6正常小鼠的粪菌悬液7d),再用解毒祛瘀滋阴方进行治疗,并用Illumina MiSeq高通量测序技术检测肠道菌群的改变,采用ELISA、RT-qPCR和HE染色评估狼疮病情变化。结果:在3种含不同初始肠道菌群的MRL/lpr小鼠中,解毒祛瘀滋阴方治疗过程中改变的肠道菌群和缓解狼疮的效果均具有差异。在正常肠道菌群的小鼠中,解毒祛瘀滋阴方能显著降低血清anti-dsDNA、IL-6、Cr和LDH的含量(P<0.01,P<0.05),降低肾脏TLR7/9基因的表达(P<0.01)和缓解肾脏病变,然而在抗生素小鼠中仅能显著降低血清anti-dsDNA和IL-6含量(P<0.01),在粪菌移植小鼠中未起到任何显著效果。结论:不同初始肠道菌群可影响解毒祛瘀滋阴方治疗SLE的疗效。Objective:To explore the effects of different initial gut microbiota on the treatment of MRL/lpr mice with Jiedu Quyu Ziyin Decoction.Methods:Female MRL/lpr lupus mice were randomly divided into model control group(intragastric administration of 0.9%sodium chloride solution for 21 days),antibiotic group(intragastric administration of antibiotic suspension for 14 days,then intragastric administration of 0.9%sodium chloride solution for 7 days)and fecal microbiota transplantation group(intragastric administration of antibiotic suspension for 14 days,then intragastric administration of fecal microbiota suspension of C57 BL/6 normal mice for 7 days).And then the MRL/lpr mice were treated with Jiedu Quyu Ziyin Decoction.Illumina MiSeq high-throughput sequencing technology was used to investigate the alterations of gut microbiota,The technique of ELISA,RT-qPCR and HE staining were used to evaluate the changes of lupus condition.Results:There were significant differences in the alterations of gut microbiota and therapeutic effects of Jiedu Quyu Ziyin Decoction in the three types of MRL/lpr mice with different initial gut microbiota.Jiedu Quyu Ziyin Decoction could significantly reduce the serum anti-dsDNA,IL-6,Cr and LDH concentration(P<0.01,P<0.05)and the expression of renal TLR7/9(P<0.01),and alleviate renal lesions in MRL/lpr mice that had not interfered with the gut microbiota.However,Jiedu Quyu Ziyin Decoction only reduced serum anti-dsDNA and IL-6 concentration in antibiotics-treated MRL/lpr mice(P<0.01)and had no therapeutic effect in fecal microbiota transplantation treated mice.Conclusion:Initial gut microbiota could affect the therapeutic effects of Jiedu Quyu Ziyin Decoction in the treatment of SLE.
