摘  要:目的:建立肝肾阴虚复合视疲劳大鼠模型,基于证效相关探讨中药复方新产品杞菊明目片滋补肝肾益精明目功效。方法:采用灌胃左甲状腺素钠溶液结合夹尾情志刺激的方法建立肝肾阴虚证候模型,同时采用15 000Lux的蓝光灯源照射复合视疲劳模型。48只SPF级SD大鼠随机分为空白组,模型组,杞菊地黄丸组、杞菊明目片高、中、低剂量组,每组8只,连续给药14d。采用酶联免疫法、比色法等测定大鼠血清环核苷酸、内分泌激素、生化水平等与肝肾阴虚证候相关指标,测定眼球组织环核苷酸和氧化因子水平,HE染色法观察大鼠视网膜形态等与视疲劳相关指标。结果:与空白组比较,模型组大鼠血清环核苷酸水平cAMP/cGMP显著升高(P<0.05),激素水平T4显著升高(P<0.01),T3显著降低(P<0.01),血清生化水平AST显著升高(P<0.05),ALB显著降低(P<0.05),免疫水平胸腺指数显著降低(P<0.05),视网膜组织中度病变。杞菊明目片各剂量组对cGMP、cAMP/cGMP、E2、E2/T、T3、CORT、ALB、TP、SOD、MDA、脾脏指数等指标有不同程度的改善(P<0.05,P<0.01),其中对血清环核苷酸、性激素、氧化水平部分指标的改善作用与阳性药杞菊地黄丸相当。结论:本研究首次创新性地建立了肝肾阴虚证复合视疲劳大鼠模型,并以经典药物杞菊地黄丸为对照,对中药复方新产品杞菊明目片功效进行评价,实验表明其可通过改善模型大鼠生物学指标发挥滋补肝肾、益精明目功效。Objective:To establish a rat model of liver-kidney yin deficiency syndrome combined with visual fatigue,and to explore the efficacy of Qiju Mingmu Tablets,a new traditional Chinese medicine compound,in nourishing liver and kidney and benefiting eyesight.Methods:The model was established by gavage of levothyroxine sodium solution combined with clip-tail emotion stimulation,and the model of combined asthenopia function was illuminated by 15000 Lux blue light source.Forty-eight SPF grade SD rats were randomly divided into a normal group,model group,Qiju Dihuang Pills group,Qiju Mingmu Tablets high,medium and low-dose groups,8 in each group,for 14 d of continuous administration.The levels of serum ring nucleotide,endocrine hormone,biochemistry related to liver-kidney yin deficiency syndrome and eye tissue ring nucleotide and oxidation factor related to visual fatigue were measured by Elisa and Colorimetry,the morphological changes of rat retina were observed by HE staining.Results:Compared with the normal group,the serum cyclic nucleotide levels of cAMP/cGMP in the model group was significantly increased(P<0.05),hormone level T4 was significantly increased(P<0.01),T3 was significantly decreased(P<0.01),and serum biochemical levels of AST was significantly increased(P<0.05),while ALB was significantly decreased(P<0.05).The immune thymus index decreased(P<0.05).Moderate retinal lesion Qiju Mingmu Tablets improved cGMP,cAMP/cGMP,E2,E2/T,T3,CORT,ALB,TP,SOD,MDA,spleen index indicators in different degrees(P<0.05,P<0.01),and the improvement of some indexes of serum cyclic nucleotide,sex hormone and oxidation level was comparable to that of positive drugs.Conclusion:This study innovatively establishes the visual fatigue of rats model in liver-kidney yin deficiency syndrome for the first time,and compares to Qiju Dihuang Pills for control,the efficacy of Qiju Mingmu Tablets is evaluated,experiment shows that it could nourish liver and kidney and benefit eyesight by improving biological indexes of model rats.
