摘  要:目的:研究砭贴预处理对低压缺氧大鼠心肌组织缺血水肿的预防作用及对心肌组织水通道蛋白-1(AQP-1)表达的影响。方法:将36只雄性SD大鼠随机分为正常组、药物组、模型组、砭贴组,每组9只。在百会、内关穴处,模型组每天贴胶布1h,砭贴组每天贴砭贴并用胶布固定1h,药物组给予红景天胶囊280mg/kg的剂量每天灌胃1次,均预处理14d。预处理完后,除正常组外,其它组均在第15天进入低压氧舱,模拟海拔6 000m高原,持续24h(断食断水),建立高原低压缺氧模型。出舱后取出心脏,HE染色观察心肌组织形态学变化;RTPCR技术检测心肌组织中AQP-1 mRNA的表达;Western blot检测心肌组织中AQP-1蛋白表达水平。结果:与正常组比较,模型组大鼠心肌组织损伤明显,AQP-1 mRNA和蛋白表达水平显著升高(P<0.05);与模型组比较,砭贴组和药物组大鼠心肌组织损伤减轻,AQP-1 mRNA和蛋白表达水平显著降低(P<0.05)。结论:砭贴预处理对急性低压缺氧大鼠心肌组织缺血水肿具有显著的预防作用,可显著降低低压缺氧状态造成的大鼠心肌组织缺血水肿程度,其机制可能与降低心肌组织中AQP-1的表达有关。Objective:To study the preventive effects of plaster acupuncture preconditioning on myocardial ischemic edema and the expression of aquaporin-1(AQP-1)in rats with hypobaric hypoxia.Methods:Thirty-six male SD rats were randomly divided into normal group,drug group,model group and plaster acupuncture group,with 9 rats in each group.At Baihui(DU 20)and Neiguan(PC 6),the model group was treated with adhesive tape for 1 h every day,the plaster acupuncture group was treated with adhesive tape for 1 h every day,and the drug group was treated with Rhodiola Capsule(280 mg/kg)by gavage once a day for 14 d.After pretreatment,except for the normal group,the other groups should enter the hypobaric oxygen chamber on the 15 th day to simulate the altitude of 6000 m for 24 h(without food or water).After the heart was taken out of the cabin,HE staining was used to observe the morphological changes of myocardial tissue;RT-PCR was used to detect the expression of AQP-1 mRNA in myocardial tissue;Western blot was used to detect the expression level of AQP-1 protein in myocardial tissue.Results:Compared with normal group,the myocardial injury of model group was obvious,and the expression levels of AQP-1 mRNA and protein were significantly increased(P<0.05);Compared with model group,the myocardial injury of plaster acupuncture group and drug group was alleviated,and the expression levels of AQP-1 mRNA and protein were significantly decreased(P<0.05).Conclusion:Plaster acupuncture preconditioning has a significant preventive effect on myocardial ischemia edema in rats with acute hypobaric hypoxia,and can significantly reduce the degree of myocardial ischemia edema in rats with hypobaric hypoxia.The mechanism may be related to the reduction of AQP-1 expression in myocardial tissue.
