摘 要:目的通过网络药理学方法探究续断-菟丝子药对对复发性流产(Recurrent spontaneous abortion,RSA)的作用机制。方法在中药系统药理学数据库与分析平台(Traditional chinese medicine systems pharmacology.database and analysis platform,TCSMP)中对续断-菟丝子药对有效活性成分进行筛选,并得到其可能的作用靶点;在GeneCards、OMIM、DrugBank、DisGeNET 4个数据库检索RSA相关靶点;利用韦恩图得到药对和疾病共同靶点;利用Cytoscape软件构建药对-成分-疾病靶点网络图;通过STRING网站完成蛋白互作网络构建;利用R软件对续断-菟丝子药对对RSA有效作用靶点进行GO与KEGG分析。结果筛选出续断-菟丝子药对有效活性成分13个,作用于178个预测靶点;筛选出疾病靶点1913个;韦恩图得到共同靶点111个;靶点涉及脂多糖反应等2010个生物过程,通过调控肿瘤坏死因子信号转导通路等152条信号通路起作用。结论研究初步揭示了续断-菟丝子药对对RSA的作用机制,为临床中药治疗RSA提供思路借鉴与理论支持。Objective To explore the mechanism of Dipsacus-Cuscuta on recurrent abortion by network pharmacology.Methods In TCSMP database,the effective active components of Dipsacus-Cuscuta were screened and its potential targets were obtained.Search RSA targets in GeneCards,OMIM,DrugBank and DisGeNET.The Venn diagram was used to obtain the common target of drug pair and disease.Cytoscape software was used to construct drug pair-component-disease target network diagram.Protein interaction network construction through STRING website was completed.The GO and KEGG were carried out on the effective target of Dipsacus-Cuscuta drug pair on RSA by Using R software.Results 13 active components of Dipsacus-Cuscuta were selected,which were applied to 178 predicted targets.1913 disease targets were screened out.The Venn diagram obtained 111 common targets.Targets are involved in 2010 biological processes such as lipopolysaccharide reaction,and play a role by regulating 152 signaling pathways such as tumor necrosis factor signaling pathway.Conclusion The study preliminarily revealed the mechanism of Dipsacus-Cuscuta on recurrent abortion,and provided a reference and theoretical support for clinical Chinese medicine treatment of RSA.
