摘 要:目的探讨德都红花-7味散在肝再生各阶段对增殖细胞核抗原(PCNA)的调节作用。方法部分肝切除术(PHT)法建立大鼠肝再生模型,按体重将模型大鼠分成模型组、阳性对照组(0.28 g·kg^(-1)护肝片)、实验组(0.6g·kg^(-1)德都红花-7味散),每组30只,灌胃,qd,连续7 d,进行PHT,后168 h各组取10只,计算肝再生率;以免疫组织化学(IHC)法检测肝组织中PCNA蛋白表达。结果模型组、阳性对照组、实验组在PHT后168 h肝再生率分别为(41.56±3.97)%,(54.09±8.95)%,(54.31±3.42)%,实验组与模型组比较,差异有统计学意义(P<0.05)。假手术组、模型组、阳性对照组、实验组在PHT术后168 h肝组织中PCNA蛋白表达为0.35±0.08,1.51±0.23,1.75±0.16,1.78±0.23,实验组与模型组比较,差异有统计学意义(P<0.05)。结论德都红花-7味散通过促进PCNA蛋白表达,从而参与了肝细胞细胞周期的调控,进而促进了肝细胞的增殖,调节了肝再生功能。Objective To investigate the regulatory effect of Deduhonghua-7 Weisan on proliferating cell nuclear antigen in liver regeneration.Methods The rat model of liver regeneration was established by partial hepatectomy(PHT),and the model rats were divided into model group,positive control group(0.28 g·kg^(-1)Hugan tablet)and experimental group(0.6 g·kg^(-1)dedu honghua-7 powder)according to body weight,with 30 rats in each group.The rats were given PHT by gavage once daily for 7 consecutive days.After 168 hours,10 rats in each group were taken to calculate the liver regeneration rate.The expression of PCNA protein in liver tissue was detected by immunohistochemistry(IHC).Results The liver regeneration rates of the model group,the positive control group and the experimental group were(41.56±3.97)%,(54.09±8.95)%,(54.31±3.42)%at 168 h after PHT,respectively.The differences between the experimental group and the model group were statistically significant(P<0.05).The expressions of PCNA protein in liver tissues of the sham group,model group,positive control group and experimental group at 168 h after PHT were 0.35±0.08,1.51±0.23,1.75±0.16,1.78±0.23.The differences between the experimental group and the model group were statistically significant(P<0.05).Conclusion Dedu Honghua-7 powder is involved in the regulation of hepatocyte cell cycle by promoting the expression of PCNA protein,thus promoting the proliferation of hepatocytes and regulating the liver regeneration function.
