摘 要:目的探讨内质网-线粒体通路中线粒体融合基因2(mitofusin 2,MFN2)的表达与肺动脉高压(pulmonary arterial hypertension,PAH)的关系。方法雄性Wistar大鼠随机分为4组:正常对照组(无任何处理),PAH组(腹腔注射60 mg/kg野百合碱诱导建立PAH模型),PRE组(PAH诱导当天开始每天使用4-PBA,持续4 w)和REV组(PAH诱导第3周每天使用4-PBA,持续2 w)。分别用BL-420F生物信息采集系统、HE染色及分子生物学手段检测4组大鼠血流动力学、病理形态学及MFN2的表达水平。结果4组大鼠平均肺动脉压力、平均右心室压力、膜厚度(thickness between internal and external elastic plates,elastic plates,elastic plates,elastic plates,WT)、血管外径(the diameter distance among external elastic plates,ED)、管腔面积(vascular lumen area,LA)、管壁面积(vascular wall area,WA)、管总面积(total vascular area,TA)、WT%、WA%和LA%比较,差异均有统计学意义(P<0.05)。4组大鼠MFN2表达水平比较,差异有统计学意义(P<0.05),其中PAH组大鼠的MFN2表达水平较REV组和PRE组大鼠低。PAH组大鼠的肺小动脉内、外弹力板的距离增宽,差异有统计学意义,管壁厚度较正常对照组增高;REV组和PRE组大鼠的肺小动脉内、外弹力板的距离比较,差异有统计学意义,PAH组大鼠的肺小动脉内、外弹力板的距离缩短,但管壁厚度与正常对照组比较,差异无统计学意义(P>0.05)。结论大鼠PAH模型中MFN2表达抑制,使血流动力学增加,肺小动脉病理形态改变及右心室肥厚指数增高。Objective To explore the correlation between the expression of MfN2 for the key factor in endoplasmic reticulum mitochondrial pathway and the pathophenotype of PAH.Methods Male Wistar rats were randomly divided into four groups:the normal control group(no treatment),the PAH group(Pulmonary hypertension in rats were developed after intraperitoneal injection administration of monocrotaline 60 mg/kg),the PRE group(the 4-PBA was used for 4 weeks from the day of PAH inductio)and the REV group(the 4-PBA was used for 2 weeks from the third week of PAH induction).The expression of MFN2 in the PERK signaling pathway,hemodynamics and pathological morphology of rats in each group was detected by the BL-420F bioinformatics acquisition system,HE staining and molecular biological methods.Results The difference of the average pulmonary artery pressure、the average right ventricular pressure、WT,ED,LA,WA,TA,WT%,WA%and LA%of the four groups was statistically significant(P<0.05).The difference of the expression levels of MFN2 in the four groups was statistically significant(P<0.05).The expression of MFN2 in PAH group were lower than those in REV group and PRE group.The normal control group:the distance between internal and external elastic plates of pulmonary arterioles was normal and the wall of pulmonary arterioles was thin;the PAH group;the distance between internal and external elastic plates of pulmonary arterioles was wider and the wall of pulmonary arterioles was thicker than that of normal control group;the REV group and the PRE group:the distance between internal and external elastic plates of pulmonary arterioles was shorter than that of PAH group and the thickness of pulmonary arterioles was not significantly different from that of normal control group difference(P>0.05).Conclu⁃sion The expression of MFN2 is inhibited in PAH model for rats,which increased hemodynamics,pathological changes of pulmonary arterioles and right ventricular hypertrophy index.
