摘  要:目的确定麦冬润肺、生津有效部位提取最佳工艺条件;通过麦冬有效部位对肺炎支原体(MP)感染小鼠的脏器指数、肺组织的病理改变结果,探讨麦冬有效部位对肺炎支原体肺炎感染小鼠肺组织炎症及免疫修复的影响。方法采用75%乙醇加热回流法提取麦冬皂苷,热水浸提法提取水溶性麦冬多糖。将100只BALB/c小鼠,随机分为10组,每组10只,分别为空白组,模型组,阿奇霉素组,养阴清肺组,麦冬多糖高、中、低剂量组,麦冬皂苷高、中、低剂量组。除空白组外其余各组采用肺炎支原体肺炎滴鼻法造模,连续造模3 d后,开始灌胃给药,每天1次,阿奇霉素连续给药5 d,其他组连续给药6 d后,进行取材,采集小鼠肺组织;采集胸腺和脾脏并立即称重,并对肺组织炎症进行病理学观察。结果按照最佳提取工艺提取麦冬总皂苷的含量为59.21%,麦冬多糖含量为64.38%;小鼠脏器指数显示,脾脏指数与模型组对比,麦冬皂苷中、低剂量组及麦冬多糖低剂量组具有极显著差异(P<0.01),阿奇霉素组、养阴清肺组及麦冬多糖中剂量组具有显著差异(P<0.05);胸腺指数与模型组对比,养阴清肺组,麦冬皂苷中、低剂量组,麦冬多糖中、低剂量组具有极显著差异(P<0.01),阿奇霉素组及麦冬皂苷高剂量组具有显著差异(P<0.05)。肺组织病理显示,与空白组比较,模型组可见明显炎症(P<0.01);与模型组比较,阿奇霉素组、养阴清肺组、麦冬多糖高、中剂量组及麦冬皂苷低剂量组的肺组织炎症明显减轻,具有极显著差异(P<0.01)。结论麦冬有效部位中总皂苷及总多糖的提取分离工艺可使有效部位总皂苷及多糖的含量均可达到50%以上,且麦冬皂苷及麦冬多糖对肺炎支原体肺炎感染小鼠有免疫调节作用,对肺组织炎症有改善作用。Objective To determine the optimal technological conditions for extracting the effective parts of dwarf lilyturf tuber for moistening lung and promoting fluid production,and to explore its effect on inflammation and immune repair of lung tissues in mice infected with Mycoplasma pneumoniae(MP)through the results of organ index and pathological changes of lung tissues.Methods In this experiment,saponins were extracted from dwarf lilyturf tuber by 75%ethanol heating reflux method,and water-soluble dwarf lilyturf tuber polysaccharides were extracted by hot water extraction method.100 BALB/c mice were randomly divided into 10 groups,with 10 mice in each group,namely blank group,model group,azithromycin group,Yangyin Qingfei group,dwarf lilyturf tuber polysaccharide high,medium and low dose groups,and dwarf lilyturf tuber saponin high,medium and low dose groups.Except the blank group,the remaining groups were modeled by nasal drip method of Mycoplasma pneumoniae pneumonia.After continuous modeling for 3 days,the mice were given azithromycin by gavage once a day for 5 days.After 6 days of continuous administration in the other groups,samples were taken and lung tissues were collected.Thymus and spleen were also collected and weighed immediately,and pathological observation of lung tissue inflammation was carried out.Results According to the best extraction process,the content of total saponins and polysaccharides in dwarf lilyturf tuber was 59.21%and 64.38%respectively.The viscera index of mice showed that compared with the model group,spleen index of ophiopogonin medium and low dose groups and ophiopogonis polysaccharide low dose group had extremely significant differences(P<0.01),and there were significant differences in the azithromycin group,Yangyin Qingfei group and ophiopogonis polysaccharide medium dose group(P<0.05).Compared with the model group,thymus index of Yangyin Qingfei group,ophiopogonin medium and low dose groups,ophiopogonis polysaccharide medium and low dose groups had extremely significant difference
