摘 要:目的探讨羊栖菜多糖(Sargassum fusiforme polysaccha-ride,SFPS)在3T3-L1细胞脂代谢中的作用。方法培养3T3-L1细胞,并建立胰岛素抵抗模型,胰岛素抵抗模型细胞经不同浓度SFPS处理48h后,测定葡萄糖消耗量(glucose consumption,Glu)、游离脂肪酸(Free fatty acid,FFA)生成量和细胞中甘油三酯(triglyceride,TG)含量。MTT法检测细胞相对增殖率,RT-PCR检测3T3-L1细胞中GLUT-4、HSL的mRNA表达情况。结果与模型组比较,不同浓度SFPS显著增加胰岛素抵抗3T3-L1细胞葡萄糖消耗量,显著降低FFA生成量,显著增加GLUT-4 mRNA 的表达,显著降低HSL mRNA的表达。结论 SFPS改善3T3-L1脂肪细胞胰岛素抵抗的分子机制可能与调控GLUT-4和HSL mRNA的表达有关。Objective To study the effect of Sargassum fusiforme polysaccharide (SFPS) on lipid metabolism of 3T3-L1 adipocytes.Methods 3T3-L1 cells were cultured and induced to insulin resistance model.The levels of glucose consumption,free fatty acid (FFA) generation and cellular triglyceride(TG) were determined after the cells treated with different concentrations of SFPS for 48 h.MTT method was used to measure cell proliferation activity and the mRNA expression of GLUT-4、HSL gene were detected by RT-PCR.Results Compared with model group,various doses of SFPS significantly promoted glucose consumption,and restrained FFA production remarkably.Compared with model group,various doses of SFPS could effectively decrease the level of HSL mRNA expression and improve the level of GLUT-4 mRNA expression.Conclusion SFPS can significantly improve IRstatus of 3T3-L1 cells probably through regulating GLUT-4 and HSL mRNA expression.
