摘 要:目的明确阴道微环境对水通道蛋白2(AQP2)mRNA在阴道上皮细胞中表达的影响。方法选取2014年5月购自美国菌种保藏中心(ATCC)的VK2/E6E7细胞作为研究对象。体外培养VK2/E6E7细胞,分别予以不同浓度的雌二醇、睾酮、孕酮、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)及不同pH的培养基干预细胞,观察并收集干预24h及48h后的细胞,采用Real Time PCR技术,明确AQP2mRNA的表达水平。结果高浓度雌二醇可增加AQP2mRNA表达水平;较低浓度孕酮增加AQP2mRNA表达水平,高浓度孕酮降低AQP2mRNA表达水平;睾酮、炎症因子(IL-6、IL-8)及pH均对AQP2mRNA表达水平无显著影响规律,但不同pH会影响阴道上皮细胞的生存状态,进而影响阴道润滑。结论阴道微环境会影响AQP2mRNA在阴道上皮细胞中的表达,雌激素缺乏可能是女性阴道润滑障碍的致病因素。Objective To investigate the effect of vaginal microenvironment on AQP2 mRNA expression in vaginal epithelial cells. Methods The VK2/E6 E7 cells purchased from American type culture collection(ATCC) in May 2014 were selected as the research objects.VK2/E6 E7 cells were cultured in vitro. The cells were pretreated with different concentrations of estradiol, testosterone, progesterone, IL-6, IL-8 and different pH values. The cells were observed and collected 24 hours and 48 hours after intervention. The expression of AQP2 mRNA was determined by real time PCR. Results High concentration of estradiol can increase the expression level of AQP2 mRNA. Low concentration of progesterone increased the expression level of AQP2 mRNA, while high concentration of progesterone decreased the expression level of AQP2 mRNA. Testosterone, inflammatory cytokines(IL-6, IL-8) and pH had no significant effect on the expression level of AQP2 mRNA, but different pH would affect the survival state of vaginal epithelial cells, and then affect vaginal lubrication. Conclusions Vaginal microenvironment can affect the expression of AQP2 mRNA in vaginal epithelial cells. Estrogen deficiency may be the pathogenic factor of vaginal lubrication disorders in women.
