摘  要:目的研究汉黄芩素对人肝癌HepG2细胞增殖、凋亡的影响及可能机制。方法常规培养HepG2细胞。MTT法检测汉黄芩素对HepG2细胞增殖的影响。流式细胞术检测经汉黄芩素处理后HepG2细胞周期、凋亡及活细胞内Caspase-3的表达。免疫印迹法检测Bcl-2和Caspase-3蛋白的表达。结果汉黄芩素6.25~50μg·mL^(-1)浓度能明显抑制HepG2细胞增殖,促进凋亡及活细胞内Caspase-3活性的表达,与对照组相比有显著性差异(P<0.05)。汉黄芩素能改变HepG2细胞周期,使S期比例逐渐减少。汉黄芩素能显著抑制Bcl-2和Caspase-3蛋白表达量。结论汉黄芩素能抑制HepG2细胞增殖,改变细胞周期,促进凋亡,其凋亡机制可能与改变Caspase-3和Bcl-2表达有关。OBJECTIVE To study the effect of wogonin on proliferation and apoptosis to HepG2 cells.METHODS HepG2 cells were cultured routinely.MTT assay was used to detect the effect of wogonin on HepG2 cells proliferation.The expressions of Caspase-3,cell cycle and apoptosiswere measured with flowcytometric analysis.The protein content of Bcl-2 and Caspase-3 were defined by Western blot.RESULTS Wogonin could obviously inhibit HepG2 cells proliferation and change the cell cycle,also could promote apoptosis and Caspase-3 expression at the concentration of 6.25-50μg·mL^(-1).With the increase of wogonin concentration,the content of Bcl-2 and Caspase-3 proteins gradually decreased.CONCLUSION Wogonin could induce the apoptosis which may be related to the expression of Caspase-3 and Bcl-2.
