摘 要:目的正交试验法优选浙江金线兰多糖最佳提取工艺。方法以料液比、提取时间、提取温度、提取次数为考察因素,采用苯酚-硫酸法测定吸光度值,以多糖提取含量为评价指标,采用L 9(34)正交试验法优选浙江金线兰多糖提取工艺。结果影响浙江金线兰多糖提取含量主次顺序为C提取温度>D提取次数>A料液比>B提取时间,提取的最佳工艺为料液比1∶30,提取时间120 min,提取温度100℃,提取次数4次,提取含量为9.95±0.23%。结论该提取工艺稳定可行,重现性好,为浙江金线兰多糖提取的最佳工艺。OBJECTIVE To optimize the best extraction process of polysaccharides from Anoectochilus zhejiangensis by orthogonal method.METHODS The ratio of material to water,extraction times,extraction temperature and extraction time as factors and absorbance value was determined by phenol-sulfuric acid method.The L 9(34)orthogonal test was used to optimize the extraction technology of polysaccharides from A.zhejiangensis.RESULTS The effect of extraction yield in descending order was extraction temperature>extraction times>ratio of material to wate>extraction time.The optimal extraction conditions polysaccharides from A.zhejiangensis were as follows:ratio of material to rate 1∶30,extraction time 120 min,extraction temperature 100℃and extraction repeated 4 times,the extraction yield was(9.95±0.23)%.CONCLUSION The optimal extraction process was stable and reproducible,which suitable for the extraction of polysaccharides from A.zhejiangensis.
