摘  要:目的建立基于SGI和DNA探针的CYP2C19*2、*3基因多态性检测的免标记荧光传感器,为CYP2C19*2/CYP2C19*3的多态性检测提供新的方法。方法应用Primer express 3.0进行CYP2C19*2、*3的两个位点的检测探针设计;建立和优化实验体系;用突变序列、错配序列对所建立方法的特异性、准确性进行评价;将所建立荧光传感体系用于模拟样本进行检测对其实际适用性进行评价。结果所建立的荧光传感体系对于CYP2C19*、*3目标序列具有较高荧光响应,且对于CYP2C19*2、*3各基因型具有不同的荧光响应,即可以准确地检测出CYP2C19*2、*3野生型、突变杂合型、突变纯合型;且所建立体系具有较较高的稳定性及可重复性。结论本研究所建立的荧光传感器具有高灵敏性、高准确性、高稳定性,可为临床CYP2C19*2、*3多态姓检测提供新思路。OBJECTIVES To design a label-free fluorescence sensor for detection of CYP2C19*2,*3 gene polymorphisms which may provide a new strategy for detection of CYP2C19*2,*3 gene polymorphisms.METHODS Apply Primer express 3.0 to design the probes for CYP2C19*2,*3 gene polymorphisms;Establish the reacting system and optimize it;Use mutational and mismatch sequences to evaluate the specificity and accuracy;Apply the fluorescence sensing system to detect of simulation to evaluate the applicability.RESULTS The fluorescence sensing system we build had a good fluorescent response to CYP2C19*2/CYP2C19*3 which could even had different responses to defferent gene type of CYP2C19*2/CYP2C19*3 target sequences,that means our fluorescence sensing system could identify the wild type,mutant heterozygote and homozygote of CYP2C19*2,*3;The fluorescence sensing system has good accuracy and stability.CONCLUSION The fluorescence sensing system exhibited excellent specificity,accuracy and stability which mayight provide a new idea for detection of CYP2C19*2,*3 gene polymorphisms in clinical practice.
