摘  要:目的运用网络药理学探讨耳聋左慈丸治疗耳聋耳鸣的作用机制。方法借助中药系统药理学数据库与分析平台(TCMSP)检索耳聋左慈丸的化学成分和相关作用靶点,同时利用Gene数据库查询与耳聋耳鸣疾病相关的靶点,从而构建出化合物-靶点网络、蛋白互作(PPI)网络、成分-靶点-信号通路网络图,研究耳聋左慈丸的作用机制。结果耳聋左慈丸处方的8味药材中筛选得出87个化合物和235个靶点,其中32个化合物未检索到相关靶点;与耳聋耳鸣疾病交集的靶点37个,PPI网络显示其中29个有相互作用关系;基因本体条目365个,前20条中包括生物过程相关条目11个,分子功能相关条目2个,细胞组成相关条目7个;京都基因与基因组百科全书通路26条,主要涉及糖尿病并发症的AGE-RAGE信号通路、FAS信号通路、PI3K-Akt信号通路和细胞外基质-受体相互作用信号通路;耳聋左慈丸处方中化合物18个(槲皮素、茵陈黄酮、山酮C排前三),相关靶点中处于前列的为钙调蛋白1(CALM1)、肿瘤坏死因子(TNF)、白细胞介素6(IL6)和酪氨酸蛋白磷酸酶非受体Ⅱ型(PTPNⅡ)。结论耳聋左慈丸处方中槲皮素、茵陈黄酮等18种活性化合物通过作用于CALM1、TNF、IL6、PTPN11等关键靶点,实现调控26条生物学信号通路,从而达到治疗耳聋耳鸣的作用。Objective To investigate the mechanism of Erlong Zuoci Pill in the treatment of deafness and tinnitus based on network pharmacology.Methods The chemical components and corresponding targets related of Erlong Zuoci Pill were searched from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The targets related to deafness and tinnitus were found through Gene database.Thus,a component-target network,the proteinprotein interaction(PPI)network,and component-target-biological pathway network diagram was constructed.Results Totally 87 components and 235 targets were obtained from 8 herbs in the Erlong Zuoci Pill through screening among which 32 compounds have no targets.37 targets related to deafness and tinnitus and the PPI network contains 29 targets.There were 365 geneontology(GO)entries,the first 20 entries including 11 biological process entries and 2 molecular function entries and 7 cellar component entries.Besides,there were 26 KEGG pathways,mainly including AGE-RAGE signaling pathway in diabetic complications,Focal adhesion,PI3K-Akt signaling pathway,ECM-receptor interaction.Conclusion The result preliminarily verifies the related mechanisms of Erlong Zuoci Pill in the treatment of deafness and tinnitus,in its prescription,18 active compounds such as quercetin,araepillin and kadsurenone can regulate 26 biological signaling pathways by acting on key targets such as CALM1,TNF,IL6 and PTPN11,so as to achieve the effect of treating deafness and tinnitus.
