摘 要:为研究阜新地区农贸市场出售的肉及肉制品产气荚膜梭菌的污染情况,并探究该菌的生物学特性。方法对2018~2020年阜新市食品风险监测项目中检出的产气荚膜梭菌进行生化鉴定和分子分型;分析了α毒素基因序列并构建了16S rRNA的系统进化树;通过药敏试验了解其耐药性。通过综合镜下形态、生化鉴定以及毛细管电泳鉴定PCR种特异性和毒素型鉴定,从2018~2020年肉及肉制品分离得到的22株菌株均为A型产气荚膜梭菌。α毒素氨基酸序列分析显示分离株C. perfringen 201901、C. perfringen 201902、C. perfringen 201905、C. perfringen 202004、C. perfringen 202006之间以及C. perfringen 201801、C. perfringen201804之间序列同源性较高,属于同一分支,亲缘关系较近。分离的菌株均对多西四环素、米诺环素和头孢氨苄的敏感性较高,对卡那霉素和庆大霉素完全耐药。本试验对当地肉及肉制品所感染的产气荚膜梭菌的生物学特性进行研究,为阜新地区肉及肉制品类食品的风险评估提供科学依据。To study the pollution condition of C.perfringens in the meat and meat products sold in farmer’s markets in Fuxin Region,and explore the biological characteristics,the method adopted in the paper is to implement biochemical identification and molecular typing for C.perfringens detected in the food risk monitoring projects in Fuxin from 2018 to 2020;analyzeαtoxin gene sequence and build 16S rRNA phylogenetic tree;and then know about the drug resistance by drug sensitivity test.Based on the form observed under comprehensive lens,the biochemical identification,and the capillary electrophoresis identification of PCR species specificity and toxin type identification,22 strains of toxin separated from the meat and meat products made from 2018 to 2020 were proved to be Type A C.perfringens.According to the analysis on the amino acid sequence ofαtoxin,sequence homology is high among the isolated strains of C.perfringen 201901,C.perfringen 201902,C.perfringen 201905,C.perfringen 202004 and C.perfringen 202006 as well as between C.perfringen 201801 and C.perfringen 201804,proving that they belong to the same branch,and are genetically related.All isolated strains are highly sensitive to doxycycline,minocyline,and cefalexin,and fully resistant to kanamycin and gentamicin.This test also studies the biological characteristics of C.perfringens infected by local meat and meat products,providing scientific basis for evaluating the risk of meat and meat products in Fuxin Region.
