摘 要:目的观察N-乙酰基半胱氨酸(NAC)抗氧化应激治疗对糖尿病小鼠骨髓干细胞增殖和凋亡的影响。方法取8~10周龄Balb/c雄性小鼠,采用链脲佐菌素腹腔注射诱导糖尿病模型。造模成功后,将小鼠分为治疗组和对照组,治疗组给予NAC腹腔注射,对照组给予生理盐水腹腔注射,连续4周。收集小鼠尿液,采用ELISA法检测8-异前列腺素(8-Isoprostane)含量;采用免疫磁珠分离法收集CD117^(+)骨髓干细胞,流式细胞术检测细胞活性氧(ROS)水平,免疫荧光分析法检测细胞增殖情况,流式细胞仪检测细胞早期凋亡情况。结果与对照组相比,治疗组尿液8-Isoprostane水平及骨髓干细胞ROS水平降低。治疗组细胞增殖能力较对照组升高,早期凋亡率较对照组降低(P<0.05或<0.01)。结论NAC可通过改善糖尿病小鼠骨髓干细胞的过氧化应激,促进骨髓干细胞增殖并减少其早期凋亡。Objective To observe the effects of N-acetylcysteine(NAC)anti-oxidant therapy on the proliferation and apoptosis of bone marrow stem cells in diabetic mice.Methods BALB/c male mice aged 8 to 10 weeks were intra⁃peritoneally injected with streptozotocin to induce the mouse models of diabetes.After the diabetes model mice were suc⁃cessfully established,we randomly divided them into the treatment group and the control group.The mice in the treatment group were given NAC by intraperitoneal injection for 4 weeks,and the mice in the control group were given normal saline(NS)by intraperitoneal injection for 4 weeks.The urine of mice was collected and the concentration of 8-Isoprostane was determined by ELISA.CD117+bone marrow stem cells were collected by the immunomagnetic bead separation.The level of reactive oxygen species(ROS)was detected by flow cytometry.The cell proliferation was detected by immunofluores⁃cence assay.Results The levels of 8-Isoprostane in the urine and ROS in the bone marrow stem cells decreased signifi⁃cantly in the treatment group as compared with those of the control group.The proliferation ability of cells increased signifi⁃cantly,and the early apoptosis rate decreased significantly in the treatment group as compared with those of the control group(P<0.05 or P<0.01).Conclusion NAC can improve the oxidative stress of bone marrow stem cells in diabet⁃ic mice,promote the proliferation and reduce the early apoptosis of bone marrow stem cells.
