摘 要:目的:初步探讨个体遗传因素对V(D)J重组过程中的偏向性影响和TCR的MHC-自身肽耐受的选择效应。方法:采用高通量测序技术分析同卵双胞胎(MZ)、异卵双胞胎(DZ)和非双胞胎(NT)外周血中TCRβ链假基因CDR3受体库与功能性CDR3受体库前后5年的动态变化。结果:TRBV基因取用,V-J配对,核苷酸的插入和删除前后5年的动态变化在MZ之间更为一致;而CDR3区TRBJ基因取用、AA取用、AA长度分布和Overlap前后5年的动态变化在MZ、DZ和NT之间无显著差异。结论:本实验提示遗传因素可能对外周血总T细胞CDR3受体库的偏向性产生影响。Objective:In order to preliminary explore the influence of individual genetic factors on bias of V(D)J recombina⁃tion and selection effect MHC-self peptide tolerance of TCR.Methods:High-throughput sequencing technology was used to analyze 5-year dynamic change of TCRβchain pseudogene and productive CDR3 repertoires among monozygotic twins(MZ),dizygotic twins(DZ),and non-twins(NT).Results:The 5-year dynamic change of TRBV gene usage,V-J pairing,and nucleotide insertion and dele⁃tion were more consistent between MZ;while the 5-year dynamic change of TRBJ gene usage,AA usage,AA length distribution and Overlap in CDR3 region did not differ significantly among MZ,DZ and NT.Conclusion:The experiment suggested that genetic factors may have a bias effect on the CDR3 repertoires of total T cells in peripheral blood.
