摘 要:蛋白质泛素化修饰包括单泛素化和多聚泛素化修饰,后者涉及结构和功能各异的8种类型的多聚泛素链,其中的线性泛素化修饰不同于由赖氨酸介导的多聚泛素化修饰,是由泛素分子以首尾相连的方式形成的一种新型多聚泛素化修饰。越来越多的研究表明,线性泛素化修饰在免疫系统中发挥重要调控作用。本文将概述线性泛素化修饰的产生、去除、识别和调节机制,并介绍其在机体免疫应答和病原免疫逃逸中的调控作用,旨在为感染和免疫性疾病防治提供新思路。Protein ubiquitination includes monoubiquitination and polyubiquitination,the latter of which involves eight structurally and functionally distinct types of polyubiquitin chains.Unlike the polyubiquitin chains formed via seven lysine residues,linear ubiquitin chain is a novel type of polyubiquitin chain linked via Met1 in a head-to-tail manner.Increasing studies have revealed the critical roles of linear ubiquitination in the regulation of the immune system.In this review,we will introduce recent research progress on linear ubiquitin chains including its generation,recognition,removal and regulatory mechanisms,with an emphasis on its regulatory roles in immune response and pathogen immune evasion,which could help provide new insights for the prevention and treatment of immune-based and infectious diseases.
