摘  要:目的阐明人参炔醇(Panaxynol,PNN)对小鼠心肌缺血再灌注损伤(MIRI)的保护作用及其可能机制。方法 30只健康雄性C57BL/6小鼠随机分成假手术组(Sham)、MIRI组和MIRI+PNN组,采用结扎冠状动脉方法建立小鼠MIRI模型,术后给予PNN干预,每日一次,连续1周。实验第7天,收集小鼠心肌组织,称取小鼠心脏重量;HE染色观察组织病理学改变;Western blot检测氧化应激和细胞凋亡相关蛋白表达。结果 MIRI组小鼠小鼠心脏重量和心脏重量指数明显升高,心肌间质出现明显的炎症细胞浸润,促氧化应激蛋白MDA5和IRE-α表达升高,而SOD-1蛋白表达降低,同时促细胞凋亡蛋白Bax和Caspase3表达升高,而Bcl-2蛋白表达降低(P<0.05)。PNN处理能显著降低MIRI诱导的小鼠心脏重量和心脏重量指数,减少心肌间质炎症细胞浸润,降低MDA5、IRE-α、Bax和Caspase3蛋白表达,提高SOD1和Bcl-2蛋白表达。此外,PNN处理还能显著增加Nrf2蛋白表达,降低ARE蛋白表达(P<0.05)。结论 NN处理对小鼠心肌缺血再灌注损伤有保护作用,其机制可能与激活Nrf2/ARE信号通路相关。Objective To elucidate the protective effect of panaxynol(PNN) on myocardial ischemiareperfusion injury(MIRI) in mice and its potential mechanism. Methods 30 healthy male C57 BL/6 mice were randomly divided into sham operation group(sham), MIRI group and MIRI + PNN group. MIRI model was established by ligation of coronary artery. PNN intervention was given once a day for 1 week. On the 7 th day of the experiment, the myocardial tissue was collected and weighed;the histopathological changes were observed by HE staining;the expression of oxidative stress and apoptosis related proteins was detected by Western blot. Results The heart weight and heart weight index of mice in MIRI group were significantly increased, the expression of Pro-oxidative stress protein MDA5 and IRE-α was increased, the expression of SOD-1 protein was decreased, the expression of Bax and caspase 3 was increased, and the expression of Bcl-2 protein was decreased(P<0.05). PNN treatment significantly reduced MIRI induced cardiac weight and cardiac weight index, decreased myocardial interstitial inflammatory cell infiltration, decreased the expression of MDA5, IRE-α, Bax and caspase 3, and increased the protein expression of SOD1 and Bcl-2. In addition, PNN treatment significantly increased expression of Nrf2 and decreased expression of ARE(P<0.05). Conclusion PNN treatment has protective effect on myocardial ischemia-reperfusion injury in mice, and the mechanism may be related to activation of Nrf2/are signaling pathway.
