摘 要:目的研究姜黄素对缺氧诱导的小胶质细胞炎症反应的抑制作用及机制。方法采用CCK8法检测姜黄素对BV2小胶质细胞的作用浓度。利用转染试剂Lipofectamine 2000进行小胶质细胞的miR-182inhibitor转染,转染后24h通过氧糖剥夺处理模拟缺血缺氧环境诱导BV-2小胶质细胞活化建立炎症模型,缺氧诱导前给予姜黄素(10μmol/L)干预2h。通过Western blot检测TLR4蛋白表达,RT-qPCR检测miR-182表达,ELISA法检测细胞培养上清液中炎症因子TNF-α、IL-6和IL-1β水平。结果姜黄素显著抑制缺氧诱导BV-2小胶质细胞分泌炎症因子TNF-α、IL-6和IL-1β水平,促进miR-182表达,抑制TLR4表达;miR-182 inhibitor可部分逆转姜黄素对缺氧诱导BV-2小胶质细胞分泌炎症因子和TLR4表达的抑制作用。结论姜黄素通过miR-182-TLR4通路抑制缺氧诱导的小胶质细胞炎症反应。Objective To study the inhibitory effect and mechanism of curcumin on hypoxia induced inflammation of microglia. Methods CCK8 assay was used to detect the concentration of curcumin on BV2 microglia. Microglia were transfected with miR-182 inhibitor by Lipofectamine 2000. 24 h after transfection, the activation of BV-2 microglia was induced by oxygen glucose deprivation(OGD) to establish the inflammatory model, and the transfected cell wastreated by curcumin for 2 hbeforeOGD. The expression of TLR4 Protein was detected by Western blot, miR-182 by RTqPCR, TNF-α, IL-6 and IL-1 β in the supernatant of cell culture were detected by ELISA. Results Curcumin significantly inhibited the secretion of TNF-α, IL-6 and IL-1 β, promoted the expression of miR-182 and inhibited the expression of TLR4. miR-182 inhibitor partially reversed the inhibition of curcumin on the secretion of inflammatory factors and TLR4 expression of BV-2 microglia induced by hypoxia. Conclusion Curcumin inhibits the inflammation of microglia induced by hypoxia via miR-182-TLR4 pathway.
