摘  要:目的探索人羊膜上皮细胞条件培养基(hAEC-CM)对使用转化生长因子β1(TGF-β1)诱导的人肝星状细胞(LX-2)迁移、增殖和Ⅰ型胶原蛋白表达水平的影响及可能的机制。方法采用Transwell迁移试验检测诱导后的LX-2细胞迁移能力的变化。MTS检测诱导后的LX-2细胞增殖能力的变化。Real-time PCR与Western blot分别检测诱导后LX-2细胞Ⅰ型胶原蛋白mRNA和蛋白表达的变化。结果人羊膜上皮细胞条件培养基显著抑制转化生长因子β1诱导的LX-2细胞迁移、增殖,显著降低Ⅰ型胶原蛋白的表达水平。结论人羊膜上皮细胞抑制肝纤维化与抑制肝星状细胞迁移、增殖和Ⅰ型胶原蛋白表达相关。Objective To investigate the effect of human amniotic epithelial cell conditioned medium(hAECCM) on the migration, proliferation and type I collagen expression of human hepatic stellate cell line(LX-2) induced by transforming growth factor β1(TGF-β1). Methods Transwell migration assay was used to detect the the migration ability of induced LX-2 cells. MTS was used to detect the proliferation ability of induced LX-2 cells. Real-time PCR and Western blot were used to detect the expression of type I collagen mRNA and protein in LX-2 cells. Results hAEC-CM could significantly inhibit the migration and proliferation of induced LX-2 cells and significantly reduce the expression level of type I collagen. Conclusion The inhibition of amniotic epithelial stem cells on the migration and proliferation of hepatic stellate cells and the expression of type Ⅰ collagen may be the reason for the inhibition of hepatic fibrosis.
