摘  要:目的探讨右美托咪定对脑缺血再灌注大鼠海马神经细胞凋亡的影响及可能机制。方法 54只雄性SD大鼠随机分为假手术组、模型组和右美托咪定处理组,线栓法制备大鼠大脑缺血再灌注模型。末次给药24 h后,检测各组大鼠脑组织含水量、伊文思蓝含量;TUNEL染色检测各组大鼠海马神经细胞凋亡;Western blot方法检测各组大鼠海马组织p-Akt、ZO-1及Claudin-5蛋白的表达。结果右美托咪定预处理能够明显降低脑缺血再灌注大鼠脑组织含水量、EB含量,减少海马组织神经细胞凋亡数,上调海马组织p-Akt、ZO-1及Claudin-5蛋白的表达(P<0.05)。结论右美托咪定能够通过抗凋亡和保护血脑屏障减轻脑缺血再灌注损伤。Objective To investigate the effect of dexmedetomidine on apoptosis of hippocampal neurons in rats with cerebral ischemia-reperfusion and its possible mechanism. Methods 54 male SD rats were randomly divided into sham group, model group and dexmedetomidine treatment group. The rat model of cerebral ischemia-reperfusion was established by suture method. 24 hours after the last administration, the water content and Evans blue content of brain tissue were detected;TUNEL staining was used to detect the apoptosis of hippocampal neurons;Western blot was used to detect the expression of p-Akt, ZO-1 and claudin-5 in hippocampus. Results Dexmedetomidine pretreatment could significantly reduce the water content and EB content of brain tissue, reduce the number of neuronal apoptosis in hippocampus, and up regulate the expression of p-Akt, ZO-1 and claudin-5 protein(P<0.05). Conclusion Dexmedetomidine can reduce cerebral ischemia-reperfusion injury through anti apoptosis and protecting blood-brain barrier.
