摘  要:目的通过对SOX10及COL2A1在先天性肺气道畸形(congenital pulmonary airway malformation,CPAM)病变组织及正常组织中的表达分析,探讨SOX10及COL2A1在CPAM的发育过程中的作用。方法2018年11月至2020年11月,中国医科大学附属盛京医院收治的30例CPAM患儿中,男19例,女11例;年龄为(44.67±37.42)个月;术前诊断为CPAM 1型17例,2型13例。收集所有患儿术中的病变肺组织及病变周围正常肺组织,将术中收集的病变肺组织作为CPAM组,将病变周围正常肺组织作为对照组。将CPAM组和对照组分别进行免疫组织化学染色、蛋白质印迹法检测及实时定量聚合酶链反应(quantificational real-time polymerase chain reaction,qRT-PCR)检测,使用t-test统计学方法对蛋白质印记法及qRT-PCR结果进行统计学分析。结果免疫组织化学染色的结果显示SOX10及COL2A1在CPAM组的表达相对于正常组织中,主要高表达于病变肺组织中的囊壁、血管壁、肺间质及异常支气管壁上,两组间的差异具有统计学意义(P<0.05)。蛋白质印迹法及qRT-PCR检测结果显示病变肺组织中SOX10及COL2A1的表达量明显高于在对照组中的表达,差异具有统计学意义(P<0.05)。结论SOX10及COL2A1在CPAM组织中存在高表达,可能参与CPAM发育过程中的囊壁、血管及异常支气管的形成过程。Objective To explore the roles of SOX10 and COL2A1 in the development of congenital pulmonary airway malformation(CPAM)by verifying their expressions in CPAM and control tissues.Methods From November 2018 to November 2020,30 CPAM children were recruited.There were 19 boys and 11 girls with an age of 44.67 months.The preoperative diagnosis was CPAM typeⅠ(n=11)and CPAM typeⅡ(n=19).The diseased and normal lung tissues were harvested intraoperatively.Both CPAM and control groups were subjected to immunohistochemical staining,Western blot and quantificational real-time polymerase chain reaction(qRT-PCR).And t-test method was employed for statistically analysis.Results Immunohistochemical staining indicated that the expression of SOX10/COL2A1 was higher in CPAM group than those in control group.Expression clustered on cyst wall,blood vessel wall,lung interstitium and abnormal bronchial wall in CPAM lung tissue(P<0.05).Western blot and qRT-PCR showed that the expression of SOX10/COL2A1 was significantly higher in CPAM lung tissue than that in control group(P<0.05).Conclusions Highly expressed in CPAM tissues,SOX10 and COL2A1 may be involved in the formation of cyst wall,blood vessel and abnormal bronchus of CPAM.
