摘 要:目的:研究补肾祛毒汤对腺嘌呤致大鼠慢性肾功能衰竭的治疗效果及其可能的分子作用机制。方法:通过对大鼠进行腺嘌呤灌胃建立慢性肾衰竭模型,造模成功后对大鼠进行给药治疗,分别给药补肾祛毒汤、补肾祛毒汤去化瘀消癥、尿毒清、贝那普利;检测各组大鼠肾功能;HE染色观察各组肾脏组织损伤情况;qPCR检测各组中CollagenⅠ、CollagenⅢ和PDGFR-αmRNA的表达;免疫组化检测组织中CollagenⅠ、CollagenⅢ和PDGFR-α蛋白的表达。结果:HE染色结果显示大鼠慢性肾衰竭模型成功建立,4组给药组均有不同程度的好转,其中补肾祛毒汤(N-DA)治疗组可见少量细胞坏死,肾小球完整,有轻微萎缩,仅有少量炎性细胞浸润。QPCR结果显示CollagenⅠ、CollagenⅢ给药组N-D A、N-D B、NDQ与BNP相较于模型组明显降低,差异显著;PDGFR-α的表达模型组相较于对照组显著降低,给药组N-D A、N-D B与BNP相较于模型组显著升高,NDQ与模型组无显著差异。免疫组化结果显示4组给药组CollagenⅠ、CollagenⅢ蛋白的表达明显较模型组减少;PDGFR-α蛋白的表达4组给药组相较于模型组PDGFR-α明显增多。结论:补肾祛毒汤对治疗慢性肾衰大鼠疗效显著,可改善肾功能,减轻肾间质纤维化,起作用方式可能是通过抑制CollagenⅠ、CollagenⅢ,促进PDGFR-α的表达实现的。Objective:To study the therapeutic effect of Nourishing Kidney and Dispelling Toxin Decoction(N-D A)on adenine-induced chronic renal failure in rats and its possible molecular mechanism.Methods:Chronic renal failure model was established by adenine gavage in rats.After the successful modeling,the rats were given drugs:N-D A,N-D B,NDQ and BNP.Renal function of rats in each group was measured.The damage of kidney tissue in each group was observed by HE staining.QPCR detection between groups of CollagenⅠ,CollagenⅢand PDGFR-αmRNA expression.Immunohistochemical detection of tissue CollagenⅠ,CollagenⅢand PDGFR-αprotein expression.Results:The results of HE staining showed that the model of chronic renal failure was successfully established in rats,and all the four groups showed different degrees of improvement.Among them,the treatment group of N-D A showed a small amount of cell necrosis,glomerular integrity,slight atrophy,and only a small amount of inflammatory cell infiltration.QPCR results showed CollagenⅠ,CollagenⅢdosage groups of N-D A,N-D B,NDQ and BNP obviously decrease compared with model group,significant difference.The expression of PDGFR-αin the model group was significantly lower than that in the controlled group,N-D A and N-D B in the drug group were significantly higher than that in the model group,and NDQ was not significantly different from that in the model group.Immunohistochemical results showed that 4 groups for CollagenⅠ,CollagenⅢprotein expression compared with the model group significantly reduced.The expression of PDGFR-αprotein in the 4 groups was significantly higher than that in the model group.Conclusion:N-D A has a good effect for the treatment of chronic renal failure rats,work way is probably by inhibiting CollagenⅠ,CollagenⅢ,promote the expression of PDGFR-α.
