摘 要:目的:观察以活血通络法为指导的葛根复方对糖尿病心肌病大鼠水通道蛋白1、3(AQP1、AQP3)及血管紧张素Ⅱ(AngⅡ)的影响,并探讨其干预作用。方法:采用STZ腹腔注射进行模型大鼠制备,并将60只成模大鼠随机分为模型组(6.5g/kg·d)、达美康组(16.7mg/kg·d)、葛芪参蒌组(7.6g/kg·d)各20只,同时选取空白大鼠10只,为对照组(6.5g/kg·d)。各组依据上述剂量,以相应药物持续灌胃9w后,对比各组水通道蛋白1、3(AQP1、AQP3)及血管紧张素Ⅱ(AngⅡ)表达变化。结果:与空白组比较,模型组大鼠AQP1、AQP3平均光密度值降低,AngⅡ值明显升高,P<0.05;与模型组比较,各治疗组大鼠AQP1、AQP3平均光密度值明显升高,AngⅡ值降低P<0.05;与达美康组比较,葛根复方组大鼠AQP1平均光密度值降低P<0.05,AQP3无变化差异P>0.05,AngⅡ值降低P<0.05。结论:以活血通络法为指导的葛根复方可升高DCM大鼠AQP1、AQP3阳性表达,降低AngⅡ值,从而改善DCM大鼠症状。由此可见,中医对糖尿病心肌病的治疗机制可能是通过活血通络作用,调节AQP1、AQP3,AngⅡ表达而达到治疗目的。Objective:Observing the effect of Pueraria Compound on aquaporin 1,3(AQP1,AQP3)and angiotensinⅡ(AngⅡ)for rats with diabetic cardiomyopathy,based on the theory of promoting blood circulation for removing obstruction in collaterals,and exploring its mechanism.Methods:STZ intraperitoneal injection was used to obtain the model rats.These model rats were then randomly divided into three groups,including model group(6.5 g/kg·d),gliclazide group(16.7 mg/kg·d)and Geqi Shenlou group(7.6 g/kg·d),with 20 rats in each group.Moreover,10 rats were selected as blank group which was used for comparison.According to the above dosage,rats in each group took medicine by gavage.After 9 weeks,the changes in aquaporin 1,aquaporin 3(AQP1,AQP3)and angiotensinⅡ(AngⅡ)in each group were compared.Results:Compared with the blank group,the average optical density values of AQP1 and AQ3 decreased and AngⅡvalue increased significantly in the model group,P<0.05.Compared with the model group,the average optical density values of AQP1 and AQ3 increased and AngⅡvalue decreased in treatment group,P<0.05.Compared with the gliclazide group,the average optical density values of AQP1 decreased,P<0.05,and that of AQP3 had no obvious difference in Geqi Shenlou group,P>0.05.Besides,AngⅡvalue decreased,P<0.05.Conclusion:Pueraria Compound guided by the theory of promoting blood circulation for removing obstruction in collaterals can improve the symptoms of rats with DCM.Meanwhile,the positive expression of AQP1 and AQ3 can be increased and AngⅡcan be decreased.It can be seen that the therapeutic mechanism may regulate the expression of AQP1,3 and AngⅡby activating blood and dredging collaterals.
