摘  要:目的:通过代谢组学技术分析脑梗死及头部电针干预对脑梗死大鼠尿液代谢组学的影响,探究电针治疗脑梗死的差异代谢标志物及关键代谢通路。方法:将36只健康成年雄性SD大鼠随机分为空白组、模型组和电针组。电针组取大鼠“百会”“前神聪”穴。各组均在术后1 d、3 d、5 d、7 d和14 d用Ludmila Belayev12分评分法评价大鼠的神经缺损。采用UPLC-Q-TOF-MS技术分析电针对MCAO大鼠尿液代谢组学的影响,进而探究电针治疗脑梗死的作用机制。结果:造模后各时间点,与空白组相比,模型组和电针组评分均显著升高(P<0.01)。随着模型组缺血时间的延长,评分逐渐下降。造模后1 d、3 d,与模型组相比,电针组评分无显著改变(P>0.05);造模后5 d、7 d和14 d,与模型组相比,电针组评分显著下降(P<0.01)。与空白组比较,模型组大鼠尿液中的乳酸、苹果酸、苯丙氨酸、鞘磷脂与磷酸胆碱等代谢物含量发生改变;与模型组相比,电针组上述代谢物发生回调。结论:电针对脑梗死大鼠的神经功能恢复具有明显的促进作用,其机制可能与电针调控三羧酸循环、氨基酸代谢和花生四烯酸代谢等途径有关。Objective:To analyze the effect of electro-acupuncture on urine metabonomics in rats with cerebral infarction based on metabonomics technology,and to explore the differential metabolic markers and key metabolic pathways of electro-acupuncture in the treatment of cerebral infarction.Methods:36 SD rats were randomly divided into the blank group,the model group and the electro-acupuncture(EA)group.In the EA group,Baihui and Qianshencong points were selected.The nerve defects were evaluated by Ludmila Belayev 12 score on the 1st,3rd,5th,7th and 14th day after the operation.UPLC-Q-TOF-MS was used to analyze the effect of electro-acupuncture on urine metabonomics in MCAO rats,so as to explore the mechanism of electro-acupuncture in the treatment of cerebral infarction.Results:Compared to those in the blank group,the scores of the model group and the EA group were significantly increased at each time point after modeling(P<0.01).With the prolongation of ischemia time,the scores decreased gradually in the model group.The scores of EA group had no significant changes compared to those in the model group on the 1st and 3rd day after modeling(P>0.05),but the scores of the EA group decreased significantly compared to those in the model group on the 5th,7th and 14th day after modeling(P<0.01).Compared to the blank group,the contents of lactic acid,malic acid,phenylalanine,sphingomyelin,choline phosphate and other metabolites in the urine were changed in the model group;which could be called back after the intervention of electro-acupuncture.Conclusion:Electro-acupuncture can promote the recovery of neurological function in rats with cerebral infarction.The mechanism may be related to the regulation of tricarboxylic acid cycle,amino acid metabolism and arachidonic acid metabolism.
