摘 要:拟南芥(Arabidopsis thaliana)CYP78A亚基因家族基因参与种子大小的遗传控制。对普通油茶(Camellia oleifera)转录组数据进行挖掘,从花蕾和根转录组数据中分别得到1条CYP78A10的同源基因,再从油茶基因组三代测序数据中鉴别出其基因组DNA序列,最后,得到2条油茶CYP78A10同源基因全长CDS序列,分别命名为CoCYP78A10a、CoCYP78A10b。用植物CYP78A亚基因家族基因的蛋白质序列构建进化树,结果表明,CoCYP78A10a、CoCYP78A10b与CYP78A10、GmCYP78A10在一个分支,可能参与油茶种子大小的调控。通过SSR位点信息分析,在CoCYP78A10a第1外显子5’端非翻译区、CoCYP78A10b的启动子区分别发现1个适宜进行SSR分子标记开发的多态性位点。CYP78A subfamily genes have been recognized to regulate seed size in Arabidopsis thaliana.Two CDSs homologous to CYP78A10 were obtained by mining transcriptome data from Camellia oleifera flower bud and root.Then,2 long DNA fragments containing the CDSs were extacted from the whole genome SMRT sequence data.Finally,2 full-length CDSs were identified and named CoCYP78A10a and CoCYP78A10b,respectively.Phylogenetic tree of CYP78As revealed that both CoCYP78A10a and CoCYP78A10b were distributed in same clan with CYP78A10 and GmCYP78A10,which suggested that both genes might regulate seed size in C.oleifera.Two SSR(simple sequence repeats)loci with high polymorphism were detected and could be employed to develop SSR markers.One SSR locus was located in 5’UTR region of the first exon of CoCYP78A10a,and another in promoter region of CoCYP78A10b.
