摘 要:利用戊二醛(GLU)交联剂的作用和溶胶-凝胶(Sol-Gel)固定化酶方法,采用"三明治"夹心结构的方法制得了Sol-Gel/AChE/GLU/GCE结构乙酰胆碱酯酶(AChE)传感器。通过传感器对氯化乙酰硫代胆碱的测定对其电极响应机理进行了探究,计算了米氏常数。结果表明:该传感器在p H 7.0磷酸缓冲溶液中对氯化乙酰硫代胆碱有明显的催化作用,固定在传感器上的AChE酶动力学响应良好,米氏常数12.6μmol/L。传感器45 d响应值仍保持90%,稳定性好,且对番茄碱测定的效果良好,有很好的应用前景。A new Sol-Gel/AChE/Glu/GCE sensor based on glutaraldehyde(Glu)crosslinker and Sol-Gel immobilized enzyme was prepared by sandwich structure method.The electrode response mechanism of acetylcholine chloride was studied by Sol-Gel/AChE/Glu/GCE sensor,and the Michaelis constant was calculated.The results showed that the sensor could catalyze acetylcholine chloride in pH 7.0 phosphoric acid buffer solution,and the AChE immobilized on the sensor had a good kinetic response with the Michaelis constant of 12.6μmol/L.The response value of the sensor was remained 90%after 45 days,with good stability and determination of lycopene,which showing a good application prospect.
