摘 要:目的:探讨活血祛湿药对糖尿病性脂肪肝大鼠肠道菌群的影响。方法:将60只6周龄清洁级Wistar雄性大鼠按照体重分为6组(空白组、模型组、对照组、活血组、祛湿组、全方组)。除空白组外其余各组大鼠通过腹腔注射链脲佐菌素(STZ)加高脂饲料诱导糖尿病性脂肪肝模型大鼠。对照组大鼠给予二甲双胍粉末混悬液灌胃;活血组、祛湿组、全方组大鼠给予相应中药水煎液灌胃,7周后收集粪便,取腹主动脉血。全自动生化仪检测血清中天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)高密度脂蛋白(HDL)含量;实时PCR定量检测厚壁菌、拟杆菌含量;气象色谱-质谱联用仪检测短链脂肪酸表达。结果:全方组大鼠与模型组、对照组相比,体重增长速度显著下降(P<0.05)。全方组大鼠TG浓度低于对照组(P=0.014);造模后各组大鼠TC LDL水平均升高(P<0.05)。祛湿组大鼠拟杆菌含量高于对照组(P=0.014)。空白组大鼠两种肠道菌与乙酸、丙酸、异丁酸、丁酸、戊酸、己酸呈正相关(r>0,P<0.01)。模型组大鼠厚壁菌与乙酸、丙酸、丁酸、戊酸呈负相关(r<0,P<0.01),与异丁酸、异戊酸、己酸呈正相关(r>0,P<0.01);拟杆菌与厚壁菌呈负相关(r=-0.949,P<0.01)。对照组大鼠两种肠道菌与各短链脂肪酸呈正相关(r>0,P<0.01),TC与两种肠道菌及LDL均呈正相关(r>0,P<0.01)。结论:活血袪湿药物对于糖尿病性脂肪肝大鼠有治疗作用,其可能机制是通过改变拟杆菌和厚壁菌与短链脂肪酸的代谢关系,影响TC代谢,达到治疗目的。活血祛湿药在提高拟杆菌丰度及降低TG含量上优于二甲双胍。Objective:To investigate the effect of blood-activating medicinal and dispel dampness medicinal on gut microbiota in rats with diabetic fatty liver disease.Methods:Sixty 6-week-old Wistar male rats were randomly divided into six groups according to their body weight results(blank group,model group,control group,blood-activating group,dispel dampness group,whole recipe group).Except for the blank group,other rats were intraperitoneally injected with STZ to induce type two diabetes fatty liver.The control group was given metformin powder suspension by gavage.The Blood-activating group,dispel dampness group,and the whole recipe group was given the corresponding Chinese medicine decoction by gavage,their feces were collected and abdominal aortic blood was taken after seven weeks.The serum concentrations of ALT,AST,TG,TC,HDL,and LDL were detected by an automatic biochemical analyzer.The contents of Firmicutes and Bacteroidtes were quantitatively determined by Real time PCR.The expression of short-chain fatty acids was detected by GC-MS.Results:Compared with the model group and the control group,the rate of weight gain significantly decreased in the whole recipe group(P<0.05).The TG concentration of the whole recipe group was lower than that of the control group(P=0.014);Both TC and LDL concentrations increased in each group after molding(P<0.05).The content of bacteroidetes in dispel dampness group was higher than that in the control group(P=0.014).The two intestinal bacteria in the blank group were positively correlated with acetic acid,propionic acid,isobutyric acid,butyric acid,valerate and caproic acid(r>,P<0.01).In the model group,firmicutria was negatively correlated with acetic acid,propionic acid,butyric acid and valeric acid(r<0,P<0.01),and positively correlated with iso-butyric acid,iso-valeric acid and caproic acid(r>,P<0.01).There was a negative correlation between bacteroidetes and firmicutes(r=-0.949,P<0.01).In the control group,two intestinal bacteria were positively correlated with each short-chain fat
