摘 要:目的研究疏肝清热通络方(SQT)对2型糖尿病大鼠脂肪酸合成酶及AMPK/eNOS通路的影响。方法75只大鼠随机分为对照组、模型组、SQT低、中、高剂量组,15只/组,采用高脂饮食联合链脲佐菌素建立2型糖尿病大鼠模型。SQT低、中、高剂量组分别灌胃给予大鼠10、20、30 g/kg的SQT,每日1次,连续8周,测定大鼠空腹血糖及糖化血红蛋白水平、血清游离脂肪酸(FFA)、三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)水平、肝组织脂肪酸合成酶(FAS)mRNA水平、心肌组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)水平、AMPKα1、p-AMPKα1、eNOS、p-eNOS蛋白水平及心肌组织病理变化。结果与模型组比较,SQT低、中、高剂量组大鼠空腹血糖及糖化血红蛋白水平、血清FFA、TG、TC、LDL-C水平、肝组织FAS mRNA及心肌组织MDA水平均显著降低(P<0.05),大鼠心肌SOD活性、AMPKα1、eNOS、p-eNOS/eNOS、p-AMPKα1/AMPKα1水平显著升高(P<0.05),心肌组织病理学明显改善。结论疏肝清热通络方能够明显降低2型糖尿病大鼠血糖及血脂水平,抑制脂肪酸合成酶基因的表达,激活心肌组织AMPK/eNOS信号通路,改善心肌氧化应激损伤。Objective To study the effect of Shugan Qingre Tongluo Recipe(SQT)on fatty acid synthase and AMPK/eNOS signaling pathway in type 2 diabetic rats.Methods 75 rats were randomly divided into control group,model group,SQT low-,medium-and high-dose groups,15 rats in each group.A high-fat diet combined with streptozotocin was used to establish a type 2 diabetic rat model.SQT low-,medium-and high-dose groups were respectively given 10,20,and 30 g/kg SQT by gavage,once a day for 8 weeks,and the fasting blood glucose,glycosylated hemoglobin levels and serum free fatty acids(FFA),Triacylglycerol(TG),total cholesterol(TC),low-density lipoprotein(LDL-C)level,liver fatty acid synthase(FAS)mRNA level,myocardial tissue superoxide dismutase(SOD)activity,malondioxide Aldehyde(MDA)level,AMPKα1,p-AMPKα1,eNOS,p-eNOS protein levels and myocardial tissue pathological changes were measured.Results Compared with the model group,the fasting blood glucose and glycosylated hemoglobin levels,serum FFA,TG,TC,LDL-C levels,liver tissue FAS mRNA and myocardial MDA levels were signifi cantly reduced in the SQT low-,medium-and high-dose groups(P<0.05).While the rat myocardial SOD activity,AMPKα1,eNOS,p-eNOS/eNOS,p-AMPKα1/AMPKα1 levels were signifi cantly increased(P<0.05).And the myocardial histopathology was significantly improved.Conclusion Shugan Qingre Tongluo Recipe can signifi cantly reduce the blood glucose and blood lipid levels,inhibit the gene expression of fatty acid synthase,activate the AMPK/eNOS signaling pathway in the myocardial tissues,and improve the myocardial oxidative stress damage in type 2 diabetic rats.
