益肾止血颗粒对Dextran诱发大鼠肠系膜急性微循环障碍的影响

摘　　要：目的探讨益肾止血颗粒对Dextran诱发大鼠肠系膜急性微循环障碍的改善作用。方法大鼠50只随机分成对照组,阳性对照组(肾炎康复片10.8 g/kg)和益肾止血颗粒13.5 g/kg、6.75 g/kg、3.4 g/kg 3个剂量组。试验共分5组,每组10只。灌胃给药1 h后,以10%Dextran 6 mL/kg舌静脉给药,制备急性微循环障碍模型。通过微循环显微镜和微图像计算机处理装置观察记录肠系膜注射Dextran 0、5、15、30、45、60 min的微动脉流速、微静脉流速、微动脉血管口径、微静脉血管口径、毛细血管开放数和微静脉血流状态的变化。结果在注射Dextran后5、15、30、45、60 min时,益肾止血颗粒能增快微血管血流速度,与对照组比较有显著差异性(P<0.01或P<0.05);注射Dextran后30、45、60 min,益肾止血颗粒能显著改善微血管痉挛状态,微动脉、微静脉血管口径与对照组比较均有显著差异性(P<0.01或P<0.05);注射Dextran后45、60 min,益肾止血颗粒能显著增加毛细血管开放数,毛细血管开放数与对照组比较均有显著差异性(P<0.05);益肾止血颗粒在注射Dextran后15、30、45、60 min,可以明显改善微循环血液流态,与对照组比较均有显著差异性(P<0.01或P<0.05)。结论益肾止血颗粒可以明显改善大鼠肠系膜微循环障碍。Objective To investigate the eff ect of Yishen Zhixue Granules on Dextran-induced acute mesenteric microcirculation disorders in rats.Methods A total of fifty rats were randomly divided into control group,positive control group(Shenyan Kangfu Tablets 10.8 g/kg)and three Yishen Zhixue Granules groups,with 13.5 g/kg,6.75 g/kg and 3.4 g/kg dosage respectively.The experiment was divided into 5 groups,each with 10 rats.One hour after intragastric administration,10%Dextran 6 mL/kg was administered intravenously through the tongue to establish the acute microcirculation disorder model.The changes of micro artery flow velocity,micro arteriolar flow velocity,micro arteriolar vessel diameter,capillary opening number and micro arteriolar flow state at 0,5,15,30,45 and 60 min after mesenteric injection of Dextran were observed and recorded with the microcirculation microscope and micro image computer processing device.Results Yishen Zhixue Granules increased the blood flow velocity of capillaries at 5,15,30,45,60 minutes after Dextran injection,which was signifi cantly diff erent from the control group(P<0.01 or P<0.05).After Dextran injection,at 30,45 and 60 minutes,Yishen Zhixue Granules signifi cantly improved the state of microvascular spasm and the diameters of arterioles and venules were signifi cantly diff erent from those of the control group(P<0.01 or P<0.05).At 45,60 minutes after injection of Dextran,Yishen Zhixue Granules signifi cantly increased the number of capillary openings,which were signifi cantly diff erent from those of the control group(P<0.05).Yishen Zhixue Granules,at 15,30,45 and 60 minutes after the injection of Dextran,signifi cantly improved the microcirculation blood flow,and there were signifi cant diff erences compared with the control group(P<0.01 or P<0.05).Conclusion Yishen Zhixue Granules can signifi cantly improve the mesenteric microcirculation disorders in rats.
