摘 要:研究探索转基因玉米在加工过程中内、外源基因的降解规律。在转基因玉米蛋白饲料加工过程的主要环节收集样本,使用微滴式数字PC R仪对加工过程中样品内、外源基因不同扩增片段进行定量研究。结果显示,酸浸对样品DNA的降解影响有限,但浸泡后的浆料湿磨以及蛋白分离阶段,样品DNA降解严重。试验通过引入的数学模型计算DNA损坏概率表明,内源zSSIIb和外源Cry1A(b)基因扩增的DNA数量与PCR产物的大小呈负相关,同时,Cry1A(b)基因降解概率更大。The study explore the degradation patterns of the endogenous and exogenous genes during processing of transgenic corn. Collect samples in the main stages of the transgenic maize protein feed process, and different amplified fragments of endogenous and exogenous genes were quantitatively studied using a microdrop digital PCR. The results showed that the acid immersion process had limited effect on the degradation of the sample DNA, but the sample DNA was severely degraded in the wet grinding and protein separation stages after immersion. The study indicates that calculation of DNA damage probability by the introduced mathematical model found the amount of DNA amplified by the endogenous zSSIIb and exogenous Cry1 A(b) genes is negatively correlated with the size of PCR products, while the probability of Cry1 A(b) gene degradation is greater.
