摘 要:解淀粉芽孢杆菌DSYZ(Bacillus amyloliquefaciens)是一种重要的生防菌株,其代谢产物对多种病原菌有着拮抗作用,广泛应用于大蒜、辣椒、花生等农作物的生物防治,但是自然条件下其分泌的代谢产物浓度较低并且浓度主要与培养基的组成,培养条件以及自身诱导肽有关。因此,通过响应面分析法对发酵培养基进行优化以及提高菌株活菌数成为当前的主要目标。首先利用添加不同碳源、氮源、无机盐的豆芽汁培养基对菌株进行发酵,测定菌数,筛选出最适的碳源、氮源和无机盐。进一步通过单因素试验考察不同浓度的碳源、氮源和无机盐对菌种菌数的影响。然后通过响应面分析法对菌株的发酵培养基进行优化,得到最优的培养基组合。优化后的发酵培养基中的最优碳源是玉米粉为18.87 g·L^(-1),最优无机氮源是氯化铵为1.36 g·L^(-1),最优无机盐是磷酸二氢钾为0.47 g·L^(-1),优化后培养基菌数达到2.20×10^(9)cfu·mL^(-1),是未优化前的1.53倍。Bacillus amyloliquefaciens(DSYZ)is an important Biocontrol Strain and its metabolites have an antagonistic effect on various of pathogenic bacteria,widely used in the biological control of crops,like garlic,pepper,peanut and so on.However,its metabolites have a low concentration under the natural conditions and the concentration is mainly related to the composition of the culture medium,culture conditions,and self-inducing peptide.Therefore,the main objective is to optimize the fermentation medium and increase the number of active strains by response surface analysis.Firstly,fermenting the strain by the soybean sprout juice medium which was added with different carbon,nitrogen and inorganic salts,then determining the number of bacteria to select the exact concentrations of carbon,nitrogen and inorganic salts.Furthermore,the effect of different concentrations of carbon,nitrogen and inorganic salts on the bacteria population was investigated by single factor experiment.After that,response surface analysis was used to optimize the strain fermentation medium,along with the optimal medium combination.Finally,the best carbon source of the optimized fermentation medium was corn meal(18.87 g·L^(-1)),the exact inorganic nitrogen source was ammonium chloride(1.36 g·L^(-1)),the optimal inorganic salt was potassium dihydrogen phosphate(0.47 g·L^(-1)),and the number of bacteria in the optimized medium reached 2.20×10^(9)cfu·mL^(-1),1.53 times of that before optimization.
