三联袋转移法在脐带血制备中的应用效果

摘　　要：目的探讨无菌环境下,三联袋转移法在脐带血有核细胞分离制备中的应用效果。方法回顾性分析2019年4月~2020年7月广州脐血库的脐血处理数据,按照分离方法分为传统法组(267份)和转移法组(278份)。比较采用传统二次离心法与将脐血转移至空袋后再进行离心的三联袋转移法在脐带血的制备质量和制备效率的差异,比较脐血冻存体积、红细胞去除率、总有核细胞数量和回收率、CD34阳性细胞数量及祖细胞集落数量等质量指标。结果转移法组的脐血平均冻存体积低于传统法组,祖细胞集落数量高于传统法组,差异有统计学意义(P<0.05)。两组的总有核细胞数、有核细胞回收率、红细胞去除率及CD34阳性细胞数量的比较,差异无统计学意义(P>0.05)。结论转移法步骤简便可行,无须增加成本,促进脐血制备质量的提高,具有实际应用价值。Objective To investigate the effect of tripe bag transfer method on the processing of cord blood total nucleated cells under aseptic surroundings.Methods A retrospective analysis of umbilical cord blood processing data from Guangzhou Cord Blood Bank from April 2019 to July 2020 and divided into traditional method group(267 cases)and transfer method group(278 cases)according to the separation method.The quality and efficiency of umbilical cord blood processing by the traditional secondary centrifugation method and the triple bag transfer method after transferring cord blood to empty bags and centrifuging were compared,and the quality indexes of cryopreservation volume,red blood cell removal rate,total nucleated cell number and recovery rate,CD34 positive cell number and progenitor cell colony number were compared between the two methods.Results The average cryopreservation volume of cord blood in the transfer method group was lower than that in the traditional method group,and the progenitor cell colonies in the transfer method group were higher than that in the traditional method group,and the differences were statistically significant(P<0.05).There were no significant differences in the total number of nucleated cells,the recovery rate of nucleated cells,the depletion rate of red blood cells and the number of CD34 positive cells between the two groups(P>0.05).Conclusion The tripe bag transfer method is a simple,feasible alternative for umbilical cord blood processing without more cost,which is shows a practical value for product quality improvement in routine cord blood banking.
