摘  要:真核翻译起始因子4E(eIF4E)基因pvr2与辣椒抗马铃薯Y病毒(Potato virus Y,PVY)和烟草蚀纹病毒(Tobacco etch virus,TEV)相关。为全面了解我国辣椒资源中pvr2的多样性及抗性,本文以1863份辣椒种质资源为研究对象,对pvr2进行了基因编码序列和抗性评价鉴定,结果显示有10种pvr2等位基因,其中已报道9种,1种未见报道,将其命名为pvr2^(23);此10种pvr2等位基因中,有2种为已报道感病等位基因,7种为已报道抗病等位基因。含有已报道抗性pvr2等位基因的材料共有649份,其中6份含有已报道抗PVY_(0),1,2的等位基因,1份含有已报道抗TEV的等位基因,642份材料含有已报道抗PVY_(0)的等位基因;含有新发现等位基因pvr2^(23)的材料有1份,在第1外显子的第185位碱基发生突变,导致编码苯丙氨酸的密码子(UGG)突变为终止密码子(UAG),人工接种PVY-LYE84.2(PVY_(0))和PVY-SON41(PVY_(0,1,2))后感病。利用SSR基因分型数据对含有抗性等位基因的材料进行遗传关系分析,为了实施基于遗传多样性的育种策略,选择了53份材料作为PVY和TEV抗性辣椒核心种质,包括所有pvr2等位基因和广泛的遗传背景。本研究通过对pvr2的多样性分析,明确了对PVY和TEV具有抗性的辣椒材料,为辣椒资源在抗性育种中的利用提供了依据;发现了一种pvr2新的变异位点,对该基因的抗性机制研究具有重要意义。The eukaryotic translation initiation factor 4E(eIF4E)gene pvr2 confers resistance to potato virus Y(PVY)and tobacco etch virus(TEV)in peppers.In order to fully understand the diversity and resistance of pvr2 in pepper germplasm in China,we evaluated the genotype and resistance of pvr2 in 1863 pepper accessions and identified 10 pvr2 alleles,of which nine have already been reported,and one genotype is new,which was named pvr2^(23).Among the 10 pvr2 alleles,two were reported susceptible alleles to PVY,and seven were reported resistance alleles.There are a total of 649 accessions containing reported resistance pvr2 alleles,of which six confer resistance against pathotype(0,1,2)of PVY,one confers resistance against TEV,and 642 confer resistance against pathotype(0)of PVY.One accession contained the newly discovered allele pvr2^(23),in which the 185th base of the first exon was mutated,resulting in the mutation of a phenylalanine codon(UGG)into a stop codon(UAG).This accession was susceptible to PVY isolates LYE84.2 of pathotype(0)and SON41 of pathotype(0,1,2)after mechhnical inoculation.We used SSR genotyping to analyze the genetic relationships between accessions containing resistance alleles.To implement a breeding strategy based on genetic diversity,53 accessions were selected as a core collection of PVY and TEV resistant pepper accessions including all pvr2 alleles and a wide range of genetic backgrounds.Through the diversity analysis of pvr2,this study identified the pepper germplasm that are resistant to PVY and TEV,and provided a basis for the utilization of pepper resources in resistance breeding.Moreover,the newly found allele pvr2^(23) is of great significance to the study of the resistance mechanism of pvr2.
