摘  要:采用小鼠急性毒性最大限度试验、兔皮肤刺激试验、小鼠骨髓嗜多染红细胞微核试验评价综合微酸性次氯酸溶液的安全性。对20只SPF级ICR小鼠一次灌胃给药,给药剂量为5 000 mg/kg BW,给药体积为20 mL/kg BW,观察小鼠临床表现及死亡情况,计算LD50。对新西兰白兔皮肤用药1 h、24 h、48 h和72 h观察并记录皮肤反应。将50只SPF级ICR小鼠随机分为阳性对照组、阴性对照组、高剂量组(5 000 mg/kg BW)、中剂量组(2 500 mg/kg BW)和低剂量组(1 250 mg/kg BW)5组,间隔30 h两次灌胃,灌胃体积20 mg/kg BW,第2次给药6 h,计数具有微核的细胞和正染红细胞,计算嗜多染红细胞和正染红细胞比率(PCE/NCE)。结果表明小鼠无死亡,微酸性次氯酸溶液小鼠急性径口LD50>5 000 mg/kg BW,属于实际无毒。对家兔一次完整皮肤刺激反应最高积分均值为0,刺激强度为无刺激性。与对照组相比,微酸性次氯酸高剂量、中剂量、低剂量组小鼠的微核细胞率均无显著性差异(P>0.05)。表明在规定剂量下使用微酸性次氯酸溶液受试剂量达5 000 mg/kg BW对小鼠骨髓嗜多染红细胞无致微核作用。说明在规定剂量下使用微酸性次氯酸溶液安全、无刺激。The acute maximum toxicity test in mice,skin stimulation test in rabbits and micronucleus test in polychromatic erythrocytes in bone marrow of mice were used to evaluate the safety of the solution.20 SPF ICR mice were administered intragastrically at a dose of mg·kg-1 BW and a dose of 20 mL·kg-1 BW.The skin reaction of New Zealand white rabbits was observed and recorded after 1 h,24 h,48 h and 72 h administration.Fifty SPF ICR mice were randomly divided into five groups:Positive Control Group,negative control group,high dose group(5 000 mg·kg-1 BW),middle dose group(2 500 mg·kg-1 BW) and low dose group(1 250 mg·kg-1 BW),at the second dose of 6 h,micronucleus cells and erythrocytes were counted,the PCE/NCE ratio was calculated.The results showed that the mice did not die,and the mice with slight acid hypochlorite solution had an acute caliber LD50>5 000 mg·kg-1 BW,which was a practically non-toxic product.The highest integral average value of skin stimulation response to intact rabbit was 0,and the stimulation intensity is non-stimulation.Compared with the control group,there was no significant difference in micronucleus cell rate among the high,middle and low dose groups(P>0.05).The results showed that the micro-acid hypochlorite solution with 5 000 mg·kg-1 BW dose did not induce micronucleus in polychromatic erythrocytes of mouse bone marrow.The results showed that the hypochlorite solution was safe and non-irritating.
